摘  要:目的 探讨氧浓度对肺腺癌增殖、侵袭转移的影响,评价不同氧浓度状态下FOXD3-AS1的表达情况。方法 收集2022年1月至2023年1月中国人民解放军中部战区总医院心胸外科肺腺癌手术68例患者的临床资料,术中活体组织标本采用液氮固定,用于提取RNA。人肺腺癌A549细胞株采用不同浓度的氧气培养,分别采用CCK-8法检测细胞增殖;Transwell法检测细胞的侵袭能力;采用qRT-PCT和Western-blot方法检测FOXD3-AS1基因和蛋白的表达情况。结果 肺腺癌组织中FOXD3-AS1的表达明显高于癌旁组织(P<0.05)。qRT-PCR检测结果表明肺腺癌A549细胞中FOXD3-AS1的表达也高于正常支气管上皮细胞16HBE(P<0.05)。随着氧浓度的提高,肺腺癌A549细胞的增殖侵袭能力明显受到抑制,FOXD3-AS1基因和蛋白的表达水平呈现下降趋势(P<0.05)。结论 FOXD3-AS1在肺腺癌组织中表达水平高于癌旁组织,其高表达与淋巴结转移阳性及肿瘤分期显著相关。随着氧浓度的升高,肺腺癌A549细胞增殖侵袭受到明显抑制,FOXD3-AS1基因和蛋白的表达水平降低。Objective To investigate the effect of oxygen concentration on the proliferation,invasion and metastasis of lung adenocarcinoma,and to evaluate the expression of FOXD3-AS1 under oxygen at different concentrations.Methods Clinical data of 68 patients with lung adenocarcinoma in cardiothoracic surgery of Central Hospital of Central Theater Command from January 2022 to January 2023 were collected.Intraoperative living tissue specimens were fixed with liquid nitrogen for RNA extraction.Human lung adenocarcinoma A549 cell line was cultured with oxygen at different concentrations,and the cell proliferation was detected by CCK-8 method.The invasive ability of cells was detected by Transwell method.The expressions of FOXD3-AS1 gene and protein were detected by qRT-PCT and Western-blot.Results The expression of FOXD3-AS1 in lung adenocarcinoma tissues was significantly higher than that in lung adenocarcinoma tissues(P<0.05).The expression of FOXD3-AS1 in A549 lung adenocarcinoma cells was also higher than that in normal bronchial epithelial cells(P<0.05).With the increase of oxygen concentration,the proliferation and invasion ability of lung adenocarcinoma A549 cells were significantly inhibited,and The expression of FOXD3-AS1 gene and protein showed a downward trend(P<0.05).Conclusions The expression level of FOXD3-AS1 in lung adenocarcinoma tissue is higher than that in paracancer tissue,and its high expression is significantly correlated with positive lymph node metastasis and tumor stage.With the increase of oxygen concentration,the proliferation and invasion of lung adenocarcinoma A549 cells were significantly inhibited,and the expression levels of FOXD3-AS1 gene and protein were decreased.
