摘  要:目的探讨微小RNA-557(miR-557)对人肺巨细胞癌低转移亚系95c细胞的影响及作用机制。方法将miR-557质粒(miR-557质粒组)和空载体(空载体组)转染到95c细胞中,同时设置空白对照组。采用MTT法检测细胞增殖,Transwell实验检测细胞侵袭,细胞划痕实验检测细胞迁移,实时荧光定量PCR和Western blot法分别检测MMP-2、MMP-9 mRNA和蛋白表达情况。结果三组细胞增殖抑制率随时间延长而增加(P<0.05)。与空白对照组和空载体组比较,miR-557质粒组细胞增殖抑制率增加,穿膜细胞数减少,细胞迁移率降低,MMP-2、MMP-9 mRNA和蛋白表达降低(P<0.05)。结论miR-557可有效抑制95c细胞的增殖、侵袭和迁移,其作用机制可能与下调MMP-2和MMP-9的表达有关。Objective To investigate the effect and mechanism of miR-557 on human lung cancer low metastatic 95c cells.Methods The miR-557 plasmid(group A) and empty vector(group B) were transfected into 95c cells.The blank cell was taken as the control(group C).The cell proliferation, invasion and migration were detected by MTT assay, Transwell assay and cell scratch assay, respectively.The mRNA and protein expressions of MMP-2 and MMP-9 were detected by real-time fluorescent quantitative PCR and Western blot.Results The inhibition rate of cell proliferation was increased with the extension of time(P<0.05).Compared with group B and group C,the inhibition rate of cell proliferation in group A was higher, the number of transmembrane cells was less, the rate of cell migration was lower and the mRNA and protein expressions of MMP-2 and MMP-9 were lower(P<0.05).Conclusion miR-557 can effectively inhibit the proliferation, invasion and migration of 95c cells.The mechanism for it may be related to the down-regulation of MMP-2 and MMP-9 expressions.
