摘 要:目的探讨慢性铝中毒对大鼠学习记忆功能的影响。方法将48只雄性Wistar大鼠划分成对照组和低、中、高剂量组,建立慢性铝中毒模型。用配置好的AlCl 3溶液对低、中、高剂量组进行灌胃,剂量分别为25、50、100 mg·kg-1,同时予以对照组等体积的生理盐水灌胃,连续90 d。用Morris水迷宫实验探究慢性铝中毒对大鼠学习记忆能力的影响;用苏木精/伊红(HE)和尼氏染色法处理大鼠脑组织切片,观察海马区神经细胞的形态变化;用实时定量PCR检测大鼠海马区CACNA1E、CALM、BDNF mRNA的表达变化;用蛋白免疫印迹实验检测海马区CACNA1E、CALM、BDNF蛋白的表达情况。结果Morris水迷宫结果显示,中、高剂量组的逃避潜伏期明显高于对照组(P<0.05或P<0.01),3个剂量组间比较差异无统计学意义(P>0.05)。HE和尼氏染色结果显示,对照组大鼠海马CA1和CA3区神经细胞排列有序,形态正常,结构完整。各剂量铝中毒组的细胞形态结构改变与铝暴露剂量有相关性,随着铝暴露剂量增加,细胞排列明显松散,形态由圆形/椭圆形变为扁梭形,胞质减少,染色由深变浅,尼氏小体明显变少。与对照组比较,中、高剂量组CACNA1E、CALM、BDNF mRNA和蛋白表达水平明显降低(P<0.05或P<0.01)。结论铝中毒会导致大鼠认知障碍,且可能与海马组织中的CACNA1E、CALM和BDNF的表达水平降低有关。Objective To explore the effects of chronic aluminum intoxication on learning memory function in rats.Methods Forty-eight male Wistar rats were divided into a control group and low,medium and high dose groups to establish a chronic toxicity intoxication model.The low,medium and high dose groups were gavaged with AlCl 3 solution at doses of 25,50 and 100 mg·kg-1,respectively,while the control group was gavaged with an equal volume of saline for 90 days.The effects of chronic aluminum intoxication on the learning and memory ability of rats were investigated by Morris water maze experiment;brain tissue sections of rats were processed with hematoxylin/eosin(HE)and Nysted staining to observe the morphologic changes of neuronal cells in hippocampal region;the changes of CACNA1E,CALM,and BDNF mRNA expression in hippocampal region of rats were examined by real-time quantitative PCR;and the expressions of CACNA1E,CALM,and BDNF proteins in hippocampus were examined by protein immunoblotting experiments.Results The results of Morris water maze showed that the escape latency of the middle and high dose groups was significantly higher than that of the control group(P<0.05 or P<0.01),and the difference between the three dose groups was not statistically significant(P>0.05).The results of HE and Niehlstrom staining showed that the neuronal cells of the hippocampal CA1 and CA3 regions of the control group were arranged in an orderly manner,with a normal morphology and structural integrity.The morphological and structural changes of cells in each dose of aluminum intoxication group correlated with the aluminum exposure dose:with the increase of aluminum exposure dose,the cells were significantly loosely arranged,the morphology changed from round/elliptical to flat spindle,the cytoplasm was reduced,the staining was changed from dark to light,and the Nitidulin vesicles became significantly less.Compared with the control group,CACNA1E,CALM,BDNF mRNA and protein expression levels were significantly lower in the medium and high dose
