摘 要:利用超声波、吐温-80、丙酮和甲苯等改变大黄欧文氏菌DXW-62的细胞膜通透性,研究其对异麦芽酮糖产率的影响。结果显示:90 W超声波处理1 min后,与空白对照组相比,异麦芽酮糖产率达到80.0%以上的时间由4.0 h缩短为3.0 h;添加浓度为5.0%的吐温-80处理后,异麦芽酮糖产率在2.0 h时已达到了75.40%,与空白对照组相比,异麦芽酮糖产率达到最高值的时间由4.0 h缩短为2.5 h;2.0%的丙酮处理后,在3.5 h时异麦芽酮糖产率达到最大,为83.04%。低浓度的甲苯处理DXW-62时的异麦芽酮糖产率无明显提高,高浓度的甲苯处理时异麦芽酮糖产率有明显下降。超声波和吐温-80共同处理后异麦芽酮糖产率远低于对照组,经碘化丙啶染色后显红色荧光细胞数量最多。Ultrasonic wave,tween-80,acetone,toluene,and other methods were used to change the plasma membrane permeability of Erwinia rhubarb DXW-62,so as to understand its effect on the yield of isomaltulose.The results showed that after 90 W ultrasonic treatment for 1 min,the time for isomaltolose yield to reach more than 80.0%was shortened from 4.0 h to 3.0 h compared with the control group.After the addition of tween-80 with a concentration of 5.0%,the yield of isomaltulose reached 75.40%at 2.0 h.Compared with the control group,the time to reach the maximum isomaltulose yield was shortened from 4.0 h to 2.5 h.After 2.0%acetone treatment,the yield of isomaltulose reached a maximum of 83.04%at 3.5 h.The yield of isomaltulose of DXW62 was not significantly increased under toluene treatment under a low concentration but significantly decreased under toluene treatment with a high concentration.The yield of isomaltulose after ultrasonic wave and tween-80 treatment was much lower than that of the control group,and the amount of red fluorescence cell was the highest after propyl iodide staining.
