摘 要:目的探究肺癌组织中内质网氧化物蛋白(ERO1L)、肿瘤坏死因子受体4(TNFRSF4)的表达与肺癌患者免疫功能、炎症反应因子及其预后的关系。方法选取2018年7月~2020年7月于本院进行手术治疗的108例肺癌患者,收集术中留取的癌组织和癌旁组织标本。采用qRT-PCR检测ERO1L和TNFRSF4的mRNA相对表达量;使用免疫组织化学法检测ERO1L和TNFRSF4蛋白表达情况,分析二者表达水平与患者临床病理特征的关系,采用Kaplan-Meier法分析ERO1L、TNFRSF4蛋白表达水平与患者预后的关系。肺癌患者预后生存率的影响因素采用Cox多因素分析。结果肺癌患者癌组织中ERO1L mRNA表达水平显著高于癌旁组织,TNFRSF4 mRNA表达水平显著低于癌旁组织(P<0.05);肺癌组织中ERO1L蛋白高表达率显著高于癌旁组织,TNFRSF4蛋白高表达率显著低于癌旁组织(P<0.05)。ERO1L蛋白高表达组患者CD3^(+)、CD4^(+)显著低于低表达组(P<0.05),IL-1β、IL-6、TNF-α显著高于低表达组(P<0.05);TNFRSF4蛋白高表达组患者CD3^(+)、CD4^(+)显著高于低表达组,IL-1β、IL-6、TNF-α显著低于低表达组(P<0.05)。ERO1L高表达组患者3年累积生存率显著低于低表达组(Log rankχ^(2)=6.100,P=0.014),TNFRSF4高表达组患者3年累积生存率显著高于低表达组(Log rankχ^(2)=11.296,P=0.001)。肺癌组织的低分化、淋巴结转移、TNM分期为Ⅲ-Ⅳ期、ERO1L高表达、TNFRSF4低表达是影响患者生存率的危险因素。结论肺癌组织中ERO1L、TNFRSF4表达与患者免疫功能、炎症因子以及预后具有一定关系。Objective To explore the relationship between the expression of endoplasmic reticulum oxidoreductin-1-like protein(ERO1L)and tumor necrosis factor superfamily member 4(TNFRSF4)in lung cancer tissues and the immune function,inflammatory response factors,and prognosis of lung cancer patients.Methods From July 2018 to July 2020,108 lung cancer patients who underwent surgical treatment in our hospital were collected,and cancer tissue and adjacent tissue samples were collected during the surgery.qRT-PCR was applied to detect the relative mRNA expression levels of ERO1L and TNFRSF4;an immunohistochemical method was applied to detect the expression of ERO1L and TNFRSF4 proteins.The relationship between the expression levels of the two and the clinicopathological features of the patients was analyzed,Kaplan-Meier method was applied to analyze the relationship between the expression levels of ERO1L and TNFRSF4 proteins and prognosis.The factors influencing the survival rate of lung cancer patients were analyzed by Cox multifactor analysis.Results The expression level of ERO1L mRNA in cancer tissues of lung cancer patients was higher than that in adjacent tissues,while the expression level of TNFRSF4 mRNA was lower than that in adjacent tissues(P<0.05);the high expression rate of ERO1L protein in lung cancer tissue was higher than that in adjacent tissue,while the high expression rate of TNFRSF4 protein was lower than that in adjacent tissue(P<0.05).The levels of CD3^(+)and CD4^(+)in the high expression group of ERO1L protein were obviously lower than those in the low expression group(P<0.05),while the levels of IL-1β,IL-6,and TNF-αwere higher than those in the low expression group(P<0.05);the levels of CD3^(+),CD4^(+)in the high expression group of TNFRSF4 protein were higher than those in the low expression group,while the levels of IL-1β,IL-6,and TNF-αwere lower than those in the low expression group(P<0.05).The 3-year cumulative survival rate of patients in the ERO1L high expression group was lower than that in the
