摘  要:目的研究薏苡仁的化学成分和抗氧化活性。方法运用硅胶柱色谱、ODS柱色谱以及半制备型高效液相色谱等技术进行分离纯化,并根据波谱数据鉴定化合物的结构;以维生素C为阳性药,采用1,1-二苯基-2-苦基肼(DPPH)和2,2′-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐(ABTS)自由基清除法测定部分苯丙素类化合物的抗氧化活性。结果从薏苡仁的乙酸乙酯和正丁醇部位中分离得到24个单体化合物,分别为鉴定为4-(3-hydroxy-propenyl)-phenol(1)、salidrosideⅠ(2)、对香豆酸(3)、阿魏酸(4)、1-O-阿魏酰甘油酯(5)、linusitamarin(6)、americanol A(7)、isoamericanol A(8)、9′-O-methylisoamericanol A(9)、3,3′-bisdemethylpinoresinol(10)、icariol A2(11)、圣草酚(12)、5,7-二羟基色原酮-5-O-β-D-葡萄糖苷(13)、5,7-二羟基色原酮-7-O-芸香糖苷(14)、1,2-heptanediol(15)、1,2,3-octanetriol(16)、3-hydroxyoctanoic acid(17)、9-hydroxynonanoic acid(18)、(+)-(4S)-(2E)-4-羟基-2-壬烯酸(19)、9-hydroxy-10-undecenoic acid(20)、8-hydroxyoctanoic acid methyl ester(21)、己烷酸单甘油酯(22)、trans-N-p-coumaroyl tyramine(23)、对羟基苯甲醇(24)。结论化合物1~2、5~11、13~22、24为首次从薏苡属中分离鉴定得到,化合物4、7~11表现出不同程度的抗氧化活性,其中化合物7、8、10的抗氧化作用较好。OBJECTIVE To study the chemical constituents and antioxidant activity of the seeds of Coicis Semen.METHODS Silica gel column chromatography,ODS column chromatography and semi-preparative high performance liquid chromatography were used to isolate and purify the compounds,and the structures of the compounds were identified according to the spectral data.The antioxidant activities of some phenylpropanoid compounds were determined by DPPH and ABTS free radical scavenging METHODS with vitamin C as positive drug.RESULTS Twenty-four monomer compounds were isolated from the ethyl acetate and n-butanol fractions of the seeds of Coicis Semen,which were identified as 4-(3-hydroxypropenyl)-phenol(1)、salidrosideⅠ(2),p-coumaric acid(3),ferulic acid(4),1-O-feruloylglycerol(5),linusitamarin(6),americanol A(7),isoamericanol A(8),9′-O-methylisoamericanol A(9),3,3′-bisdemethylpinoresinol(10),icariol A2(11),eriodictyol(12),5-β-D-glucosyloxy-7-hydroxychromone(13),5,7-dihydroxychromone-7-O-rutinoside(14),1,2-heptanediol(15),1,2,3-octanetriol(16),3-hydroxyoctanoic acid(17),9-hydroxynonanoic acid(18),(+)-(4S)-(2E)-4-hydroxy-2-nonenoic acid(19),9-hydroxy-10-undecenoic acid(20),8-hydroxyoctanoic acid methyl ester(21),monoglyceryl hexanoate(22),trans-N-p-coumaroyl tyramine(23)and p-hydroxybenzyl alcohol(24).CONCLUSION Compounds 1-2,5-11,13-22 and 24 are isolated and identified from Coix for the first time,compounds 4 and 7-11 show different degrees of antioxidant activity,among which compounds 7,8 and 10 have better antioxidant effect.
