摘  要:目的运用网络药理学和分子对接的方法,讨论中医药治疗高脂血症的作用机制。方法前期对高脂血症用药规律进行研究,得出治疗该病最有效的中药组合(山楂、何首乌、丹参、决明子、泽泻)和高频中药,收集中药成分靶点和疾病靶点,并求交集靶点,进行蛋白质互作网络(PPI)、基因本体论(GO)和京都基因与基因组百科全书(KEGG)分析。将关键靶点和高频中药成分的结构进行分子对接。结果该组合中含药物成分136个,成分靶点396个,疾病靶点501个,交集靶点37个,涉及63条通路。起效的关键靶点为激素敏感脂肪酶(LIPE)等,关键通路包括脂肪细胞脂溶解的调节等。高频中药与关键靶点的2889种结合方式中,1679种的结合能<-7.0 kcal/mol;其中陈皮的橙皮素-7-氧-芦丁苷与过氧化物酶体增殖激活受体γ(PAARG)的亲和力最强。结论该药物组合可能通过促进脂质代谢和抑制脂肪细胞分化来减少脂质、扩张血管,进而减少血管堵塞,以缓解高血脂对血管内皮的损伤。Objective Network pharmacology and molecular docking were used to discuss the mechanism of traditional Chinese medi-cine in the treatment of hyperlipidemia.Methods The most effective combination of Chinese medicine(Crataegi Fructus,Polygoni Multiflori Radix,Salviae Miltiorrhizae Radix et Rhizoma,Cassiae Semen,Alismatis Rhizoma)and high frequency Chinese medicines were discovered in the research of composition in treating hyperlipidemia in the early period.The components and disease targets of the most effective combination of Chinese medicine were collected and intersection targets were calculated for PPI,GO and KEGG analysis.Both structures of key targets and high frequency components of traditional Chinese medicine were docked by molecular docking.Results 136 drug components,396 component targets,501 disease targets,37 intersection targets and 63 pathways were con-tained in this combination of Chinese medicine.LIPE was included in the effective and key targets,the regulation of adipocyte lipoly-sis was included in key pathways.Binding energy of 1679 of 2889<-7.0 kcal/mol in the binding modes of high frequency Chinese medicine and key targets.Hesperetin-7-O-rutinoside from Citri Reticulatae Pericarpium has the strongest affinity with PAARG.Conclusion This combination of Chinese medicine might promote lipid metabolism and inhibit adipocyte differentiation to reduce lipid,expand blood vessels and reduce vascular blockage to alleviate the damage of hyperlipidemia on vascular endothelial.
