摘  要:目的 探讨miR-652-3p对胶质母细胞瘤(GAM)增殖和迁移的影响。方法 用miR-652-3p模拟物转染胶质母细胞系C6和U87,然后分别通过CCK8、细胞流式、EdU、Transwell和划痕实验研究miR-652-3p对细胞增殖和迁移的影响。利用实时定量聚合酶链反应(qRT-PCR)、蛋白质免疫印迹(Western Blot)和双荧光素酶报告基因实验检测miR-652-3p与Neuroplastin(Nptn)的互作关系。通过CCK8、细胞流式、EdU、Transwell和划痕实验研究Nptn对细胞增殖和迁移的影响。结果 miR-652-3p的过表达可以抑制C6和U87细胞的增殖和迁移;双荧光素酶报告基因实验结果表明miR-652-3p可以与Nptn结合,Nptn表达水平受到miR-652-3p水平的影响;干扰Nptn的表达可以抑制C6和U87细胞的增殖和迁移。结论 miR-652-3p通过靶向Nptn抑制GBM的增殖和迁移。Objective To investigate the effect of miR-652-3p on glioblastoma(GBM)cell lines.Methods GBM C6 and U87 cell lines were transfected with miR-652-3p mimic,and then the effects of miR-652-3p on cell proliferation and migration were detected by CCK8,flow cytometry,EdU,Transwell and wound-healing assays,respectively.The interaction between miR-652-3p and neuroplastin(Nptn)was detected by quantitative real-time polymerase chain reaction(qRT-PCR),Western blot and dual luciferase reporter gene assays.The effects of Nptn on cell proliferation and migration were investigated by CCK8,cytometry,EdU,Transwell and wound-healing assays.Results Overexpression of miR-652-3p inhibits the proliferation and migration of GBM cells.Nptn is a functional target of miR-652-3p.Interfering with the expression of Nptn inhibits the proliferation and migration of GBM cells.Conclusion miR-652-3p suppresses GBM proliferation and migration by targeting Nptn.
