摘 要:本试验旨在通过分析我国华南地区家禽配合饲料中柄曲霉素(STC)、赭曲霉毒素A(OTA)、玉米赤霉烯酮(ZEA)的污染情况,为真菌毒素风险评估提供数据支撑。从华南地区(广东省、海南省、广西壮族自治区)采集家禽配合饲料样品232份,经含20%水的甲醇提取,氯化钠和无水硫酸镁盐析后,加入N-丙基乙二胺(PSA)和C 18吸附杂质,上清液过膜后进行高效液相色谱串联质谱(HPLC-MS/MS)分析,通过基质标准曲线进行定量。分别采用卡方检验和非参数检验比较不同地区家禽配合饲料中STC、OTA、ZEA的检出率和含量。结果显示:3种真菌毒素在各自线性范围内线性关系良好,相关系数≥0.99,检出限和定量限分别为0.02~0.18μg/kg和0.07~0.59μg/kg,在低、中、高3个加标水平下,3种真菌毒素的平均回收率为83.0%~98.9%,精密度为0.8%~8.2%,日间差和日内差均<20%。232份家禽配合饲料样品中,ZEA检出率最高(100.0%),其次为STC(70.3%)、OTA(22.0%)。其中,海南省样品中STC的检出率显著高于其他地区(P<0.05)。本试验开发的方法操作简单、灵敏度高,适用于饲料中ZEA、OTA和STC的同时检测分析;我国华南地区家禽配合饲料尽管总体污染水平较低,但真菌毒素污染情况普遍,应引起重视。The aim of this study was to provide data support for the risk assessment of mycotoxins by analyzing the contamination of sterigmatocystin(STC),ochratoxin A(OTA)and zearalenone(ZEA)in poultry compound feeds from South China.A total of 232 poultry compound feeds were collected from South China(Guangdong Province,Hainan Province,Guangxi Zhuang Autonomous Region).The samples were extracted with methanol containing 20%water.After salted out with sodium chloride and anhydrous magnesium sulfate,2-propylaminoethylamine(PSA)and C18 were added to adsorb impurities.The supernatant was filtrated and analyzed by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).Accurate quantification was achieved by matrix-matched calibration curves.Chi-square test and nonparametric test were utilized to compare the detection rate and contaminant concentration of STC,OTA and ZEA in poultry compound feeds from different regions.The results showed that good linear relationships were obtained in respective linear ranges and the correlation coefficients were greater than 0.99.The limit of detection ranged from 0.02 to 0.18μg/kg and the limit of quantification ranged from 0.07 to 0.59μg/kg,respectively.Recoveries of 3 mycotoxins at low,medium and high spiked levels were ranged from 83.0%to 98.9%.Precisions were ranged from 0.8%to 8.2%,with intra-day precision and inter-day precision less than 20%.Among 232 poultry compound feed samples,ZEA had the highest detection rate(100.0%),followed by STC(70.3%)and OTA(22.0%).The STC detection rate of sample from Hainan Provine was significantly higher than that from other regions(P<0.05).In conclusion,the method developed in this experiment is simple,sensitivity and suitable for the simultaneous detection and analysis of mycotoxins in feeds.Although the overall contamination level is low,the mycotoxins pollution of feed in South China is widespread,that should be pay attention to.
