摘 要:目的探讨细胞焦亡在急性脑梗死缺血再灌注损伤中的作用和机制。方法2020年12月至2021年3月将12只SD雄性大鼠随机分为实验组和假手术组,实验组采用改良Longa法制备急性脑缺血再灌注大鼠模型,假手术组只暴露颈内动脉不阻断血流;实验组缺血处理2 h后再灌注,3 h后两组同时进行免疫荧光和蛋白质印迹(Western blot,WB)检测,最后进行脑含水量测定,同时采用酶联免疫吸附(ELISA)检测血清中IL-18和IL-1β的含量。结果实验组大鼠缺血2 h后按Bederson方法评定神经功能缺损,结果显示6只大鼠均造模成功,红四氮唑染色(TTC)提示实验组大鼠脑梗死重量百分比显著高于假手术组(P<0.001);WB检测发现实验组NLRP3、GSDMD及Caspase-1表达与假手术组差异无统计学意义;免疫荧光显示两组GFAP、Nestin和NeuN的表达均差异无统计学意义,但模型4、5、6的GFAP的表达量高于对照组;实验组脑含水量明显高于假手术组(P<0.01);实验组血清中IL-1β、IL-18的含量显著高于假手术组(P<0.001)。结论缺血早期再灌注后血清炎症因子明显增高,但脑细胞并未发现明显细胞焦亡。Objective To investigate the role and mechanism of cell charring in acute cerebral infarction with ischemia reperfusion injury.Methods During the period from December 2020 to March 2021,12 male SD rats were randomly divided into an experimental group and a sham operation group.The experimental group adopted a modified longa method to establish a rat model of acute cerebral ischemia/reperfusion.The sham operation group only exposed the internal carotid artery without blocking blood flow;The experimental group was subjected to ischemia for 2 hours before reperfusion.After 3 hours,both groups were simultaneously tested with immunofluorescence and Western blot(WB).Finally,brain water content was measured,and serum IL-18 and IL-1βwere detected by enzyme linked immunosorbent assay(ELISA)Content.Results After 2 hours of ischemia in the experimental group,neurological deficits were evaluated using the Bederson method.The results showed that all 6 rats were successfully modeled.Red tetrazolium staining(TTC)suggested that the weight percentage of cerebral infarction in the experimental group was significantly higher than that in the sham operation group(P<0.001);WB assay showed that there was no significant difference in the expression of NLRP3,GSDMD,and Caspase-1 between the experimental group and the sham operation group;Immunofluorescence showed that there was no significant difference in the expression of GFAP,Nestin,and NeuN between the two groups,but the expression of GFAP in models 4,5,and 6 was higher than that in the control group;The brain water content in the experimental group was significantly higher than that in the sham operation group(P<0.01);Experimental group serum IL-1β、The content of IL-18 in the sham operation group was significantly higher than that in the sham operation group(P<0.001).Conclusion Serum inflammatory factors significantly increase after early ischemia reperfusion,but there is no significant cell death in brain cells.
