摘 要:脏器纤维化是器官慢性炎症过程中常见的病理改变,主要特征为细胞外基质过度沉积引起组织损伤,形成永久性瘢痕,导致器官功能障碍,目前该类疾病治疗手段有限,预后较差。表观遗传学参与了纤维化进程,其中RNA N^(6)-甲基腺苷(m^(6)A)甲基化修饰作为真核生物中最常见的RNA转录后修饰通过参与信使RNA核输出、剪接、翻译和稳定等调控基因表达,从而影响生物学功能。m^(6)A甲基化修饰通过关键修饰酶调控相关通路参与脏器纤维化的形成和发展,这为脏器纤维化的治疗提供了新方向。Organ fibrosis is a common pathological change of the organs in the process of chronic inflammation,the main characteristic of which is the organ dysfunction caused by permanent scar tissue damage from the excessive extracellular matrix accumulation.At present treatment for this kind of disease is limited,and the prognosis is poor.Epigenetics is involved in the process of fibrosis,among which RNA N^(6)-methyladenosine(m^(6)A)methylation,as the most common RNA post-transcriptional modification in eukaryotes,regulates gene expression by participating in mRNA nuclear export,splicing,translation,and stability,thereby affecting the biological functions.The m^(6)A methylation participates in the formation and development of organ fibrosis by regulating related pathways through key modifying enzymes,providing a new direction for the treatment of organ fibrosis.
