摘  要:目的旨在对1,2-辛二醇的毒性与抑菌性进行考察,探讨1,2-辛二醇单独使用作为日用化妆品防腐剂的安全性与合理性。方法本研究采用长期经皮毒性试验、细胞毒性试验、胶原蛋白浓度测定试验以及抑菌效力试验在不同剂量下对受试物进行了综合考察。结果1,2-辛二醇在日均1 mg/g的剂量下长期经皮接触吸收,未见明显毒性;其对大肠埃希菌和白色念珠菌的抑菌作用浓度8 mg/ml,以及对金黄色葡萄球菌、铜绿假单胞菌和黑曲霉抑菌作用浓度16 mg/ml;受试物1,2-辛二醇在培养基中的添加浓度为2.5 mg/ml时(该剂量组与溶剂对照组比较,采用单因素方差分析;假定等方差,F=7.4,P<0.05;非假定等方差:T=6.6,P<0.001),对皮肤成纤维(HSF)细胞合成人I型胶原蛋白(Col-I)的能力具有轻微抑制作用。结论1,2-辛二醇具有较高的安全性,与较好的抑菌性,但进入真皮层中的浓度若达到2.5 mg/ml时或具有局部干扰细胞功能性蛋白合成代谢的作用。Objective To investigate the toxicity and bacteriostasis of 1,2-octanediol,and to explore the safety and rationality of 1,2-octanediol used alone as a preservative for daily cosmetics.Methods In this study,long-term transdermal toxicity test,cytotoxicity test,collagen concentration determination test(Elisa)and bacteriostatic efficacy test were used to comprehensively investigate the test substances at different doses.Results Long-term percutaneous absorption at a dose of 1 mg/g of 1,2-octanediol has no obvious toxicity.And its MIC on Escherichia coli and Candida albicans were 8 mg/ml,on Staphylococcus aureus,Pseudomonas aeruginosa and Aspergillus niger were 16 mg/ml respectively.But at the cellular level:When the concentration of the test substance added to the medium was 2.5 mg/ml(making statistics by One-Way ANOVA,comparing with solvent control group;Suppose the variances are equal,F=7.4,P<0.05;Suppose the variances are not equal,T=6.6,P<0.001),it had a slight negative effect on the secretion of human type I collagen(Col-I)by skin fibroblast(HSF).Conclusion 1,2-octanediol has high safety and good antibacterial activity,but when the concentration in the dermis reaches 2.5 mg/ml,it may have the effect of partially interfering with the synthesis and metabolism of functional proteins in cells.
