Ang-2、LYVE-1的表达与皮肤鳞状细胞癌生物学特性的相关性

摘  要:目的探究血管生成素2(angiopoietin-2,Ang-2)及淋巴管内皮透明质酸受体-1(lymphatic vessel endothelial hyaluronan receptor-1,LYVE-1)的表达与皮肤鳞状细胞癌(cutaneous squamous cell carcinoma,cSCC)生物学特性的相关性。方法收集2013年9月~2021年2月江苏大学附属医院收治的44例原发性cSCC患者,其cSCC标本中Ang-2、LYVE-1的表达情况用免疫组织化学(immunohistochemistry,IHC)方法检测,其中LYVE-1的表达情况以微淋巴管密度(microlymphatic vessel density,MLVD)表示。结果Ang-2在cSCC无淋巴结转移的组别中,高表达率为44%(15/34),明显低于淋巴结转移组的90%(9/10),差异有统计学意义(P<0.05);对于cSCC高分化组,Ang-2的高表达率为35%(9/26),明显低于中低分化组的83%(15/18),差异有统计学意义(P<0.05)。LYVE-1在cSCC无淋巴结转移组的表达为8.49±4.26,明显低于淋巴结转移组(13.48±5.91),差异有统计学意义(P<0.05);LYVE-1在cSCC高分化组的表达为6.39±2.09,明显低于中低分化组(14.28±4.42),差异有统计学意义(P>0.05)。cSCC患者的性别、年龄和肿瘤直径对Ang-2和LYVE-1的表达无影响(P>0.05)。cSCC患者的Ang-2表达水平与LYVE-1呈正相关(r=0.598,P<0.01)。结论Ang-2与LYVE-1参与推动cSCC的发生、发展以及淋巴结转移过程。Objective To investigate the correlation between the expression of angiopoietin-2(Ang-2),lymphatic vessel endothelial hyaluronan receptor-1(LYVE-1)and the biological characteristics of cutaneous squamous cell carcinoma.Methods A total of 44 patients with primary cSCC admitted to the Affiliated Hospital of Jiangsu University from September 2013 and February 2021 were included.The expression levels of Ang-2 and LYVE-1 in their cSCC specimens was detected by immunohistochemistry(IHC).The expression level of LYVE-1 was expressed by the microlymphatic vessel density(MLVD).Results The high expression rate of Ang-2 in cSCC group without lymph node metastasis was 44%(15/34),which was significantly lower than that in CSCC group(90%,9/10),and the difference was statistically significant(P<0.05).The high expression rate of Ang-2 in the highly differentiated cSCC group was 35%(9/26),which was significantly lower than that in the medium and poorly differentiated CSCC group(83%,15/18),and the difference was statistically significant(P<0.05).The expression of LYVE-1 in cSCC group without lymph node metastasis was 8.49±4.26,which was significantly lower than that in cSCC group(13.48±5.91),and the difference was statistically significant(P<0.05).The expression of LYVE-1 in highly differentiated cSCC group was 6.39±2.09,which was significantly lower than that in medium and poorly differentiated cSCC group(14.28±4.42),and the difference was statistically significant(P<0.05).The expression of Ang-2 and LYVE-1 was not related to gender,age and tumor size(P>0.05).The expression level of Ang-2 was positively correlated with LYVE-1(r=0.598,P<0.01).Conclusion Ang-2 and LYVE-1 are involved in promoting the occurrence,development and lymph node metastasis of cSCC.
