摘 要:超氧化物歧化酶(superoxide dismutase, SOD)是抗氧化系统中的核心酶。本研究通过生物信息学分析预测了草菇sod基因编码蛋白的理化性质、保守结构域和进化关系,并测定了草菇继代培养过程中的菌丝生理性状和sod基因的相对表达量。结果表明,草菇基因组中含有1个Cu/Zn-sod基因和3个Mn-sod基因,其编码蛋白均具有SOD的典型特征,属于SOD家族成员,并且草菇4个SOD分别与不同真菌的SOD具有较高的同源性。随着草菇继代培养次数的增加,菌落直径和菌丝生长速度都呈现先增加后降低的趋势;Cu/Zn-sod和Mn-sod3基因的相对表达量在继代过程中呈现先增加后降低的趋势,Mn-sod1基因在继代菌株的相对表达量均高于T0,而Mn-sod2均低于T0。本研究为揭示草菇sod基因的生物学功能和菌种继代退化机制研究奠定了基础。Superoxide dismutase is the core enzyme in the antioxidant system.In this study,the physicochemical properties,conserved structural domains and evolutionary relationships of the protein encoded by the sod gene of Volvariella volvacea were predicted by bioinformatics analysis,and the physiological characters of mycelium and the relative expression of sod genes during the subculture of Volvariella volvacea were determined.The results showed that the genome of Volvariella volvacea contained one Cu/Zn-sod gene and three Mn-sod genes,and all of them encoded proteins with typical characteristics of SOD,belonging to the SOD family members.The four SODs of Volvariella volvacea had high homology with SODs of different fungi.The relative expressions of Cu/Zn-sod and Mn-sod3 genes increased first and then decreased during subculture.The relative expressions of Mn-sod1 gene were higher than T0,while those of Mn-sod2 were lower than T0.This study lays the foundation for further revealing the biological function of sod gene in Volvariella volvacea and the mechanism of subculture degeneration of the strain.
