摘 要:以通关藤为原料,采用超声辅助水提法提取通关藤多糖,研究其结构表征及体外抗氧化、抗肿瘤细胞增殖作用。采用紫外可见分光光度计、离子色谱仪和红外光谱仪对通关藤多糖的结构进行表征,通过自由基清除能力评价通关藤多糖体外抗氧化活性。利用四甲基偶氮唑盐(MTT)法测定不同浓度通关藤多糖分别作用于人肝癌细胞HepG224 h、48 h的细胞抑制率。结果显示通关藤多糖的多糖含量为72.83%,由Glc、Gal、Ara、Rha、Xyl、Man、ManA、GalA和GlcA组成,其摩尔比为58.5∶17.8∶17.1∶2.4∶1.2∶0.9∶0.9∶0.6∶0.5;通关藤多糖对DPPH自由基、ABTS自由基和羟自由基都具有一定的清除作用,且呈现浓度依赖性。通关藤多糖可抑制人肝癌细胞HepG2增殖,且呈浓度与时间依赖性。因此,通关藤多糖具有良好的体外抗氧化和抗肿瘤细胞增殖作用,具有潜在的开发应用价值。Using Marsdenia tenacissima as raw material,ultrasonic-assisted water extraction technology was used to extract the polysaccharide from Marsdenia tenacissima,its structure characterization,antioxidant and antiproliferation effect on tumor cells in vitro were studied.The structure of the polysaccharides from Marsdenia tenacissima was characterized by UV visible spectrophotometer(UV),ion chromatography(IC),and infrared spectroscopy(IR).The scavenging rates of the polysaccharides on free radicals were evaluated.Tetramethylazolamide salt(MTT)assay was used to detect the inhibition rates of human liver cancer cells HepG2 treated with polysaccharides from Marsdenia tenacissima for 24 h and 48 h.The results showed that the polysaccharide content from Marsdenia tenacissima was 72.83%,mainly composed of glucose,galactose,arabinose,rhamnose,xylose,mannose,mannuronic acid,galacturonic acid and glucuronic acid,and the molar ratio was 58.5∶17.8∶17.1∶2.4∶1.2∶0.9∶0.9∶0.6∶0.5,polysaccharides of Marsdenia tenacissima could scavenge DPPH,ABTS and hydroxyl free radicals and exhibited concentration dependence.Polysaccharides from Marsdenia tenacissima could inhibit the growth of human liver cancer cells HepG2 in concentration and time dependent manners.The conclusion was that the polysaccharides from Marsdenia tenacissima had good antioxidant and antiproliferation effect on tumor cells in vitro.The polysaccharides from Marsdenia tenacissima has great potential value in developments and applications.
