不同剂量异丙肾上腺素对家兔动脉血压及心脏损伤的影响探讨

摘  要:目的:探讨与分析不同剂量异丙肾上腺素对家兔动脉血压及心脏损伤的影响。方法:研究时间为2022年4月—2023年2月,健康成年雄性家兔18只随机平分为低剂量异丙肾上腺素组、高剂量异丙肾上腺素组与对照组,每组6只。低剂量异丙肾上腺素组、高剂量异丙肾上腺素组家兔每日背部皮下注射0.1ml/kg与0.4ml/kg的异丙肾上腺素100μl,1次/d;对照组在同样位置注射无菌生理盐水100μl,1次/d,持续应用14d,检测家兔中心动脉压变化情况。结果:低剂量、高剂量异丙肾上腺素组处理第7天、第14天的平均动脉压、血清肌酸激酶、乳酸脱氢酶含量都高于对照组(P<0.05),高剂量异丙肾上腺素组也高于低剂量异丙肾上腺素组(P<0.05)。低剂量、高剂量异丙肾上腺素组处理第7天、第14天的维持窦性节律时间均短于对照组(P<0.05),且高剂量异丙肾上腺素组短于低剂量异丙肾上腺素组(P<0.05)。低剂量、高剂量异丙肾上腺素组处理第14天的心脏组织Caspase-3、Bcl-2蛋白相对表达水平高于对照组(P<0.05),且高剂量异丙肾上腺素组高于低剂量异丙肾上腺素组(P<0.05)。结论:高低剂量的异丙肾上腺素均可降低家兔维持窦性节律时间,提高血清肌酸激酶、乳酸脱氢酶含量,促进其心脏组织Caspase-3、Bcl-2蛋白的表达,且存在量效关系,这为异丙肾上腺素诱导家兔心肌损伤模型的构建提供机制及理论依据指导。Objective:To explore and analysis the effects of different doses of isoproterenol on arterial blood pressure in rabbits.Methods:The study period were from April 2022 to February 2023,and 18 cases of healthy adult male rabbits were equally randomly divided into low dose isoproterenol group,high dose isoproterenol group,and control group,with 6 rabbits in each group.The low dose isoproterenol group and high dose isoproterenol group rabbits were subcutaneously injected with 0.1 ml/kg and 0.4ml/kg of isoproterenol 100μl once a day.The control group were injected with sterile physiological saline at the same location for 100μl once a day,continuously applied for 14 days,to detect changes in central arterial pressure in rabbits.Results:Mean arterial pressure,serum creatine kinase,and lactate dehydrogenase levels were higher in the low-dose and high-dose isoprenaline groups than in the control group on the 7th and 14th day of treatment(P<0.05),and were also higher in the high-dose isoprenaline group than in the low-dose isoprenaline group(P<0.05).The time to maintain sinus rhythm on the 7th and 14th day of treatment was lower in the low-dose and high-dose isoprenaline groups than in the control group(P<0.05),and lower in the high-dose isoprenaline group than in the low-dose isoprenaline group(P<0.05).The relative expression levels of Caspase-3 and Bcl-2 proteins in cardiac tissues of low-dose and high-dose isoproterenol groups on the 14th day of treatment were higher than those of the control group(P<0.05),and the high-dose isoproterenol group showed a significant increase compared with the low-dose group(P<0.05).Conclusion:High and low doses of isoprenaline can reduce the time to maintain sinus rhythm,increase serum creatine kinase and lactate dehydrogenase levels,and promote the expression of Caspase-3 and Bcl-2 proteins in cardiac tissues of rabbits,and there is a quantitative relationship between them,which provides a mechanism and theoretical basis to guide the construction of isoprenaline-induced rabbit cardiac
