摘 要:目的构建小鼠下肢股动脉粥样硬化模型。方法选择8周龄载脂蛋白E敲除(ApoE-/-)的C57BL/6雄性小鼠16只,购自北京华阜康生物科技股份有限公司,采用随机数字表法将小鼠分为股动脉袖带置入组(对照组,8只)和股动脉袖带置入+串联结扎组(模型组,8只)。两组小鼠先高脂饮食诱导4周,然后分别行手术干预,再继续高脂饮食8周,实验周期总计12周。利用激光多普勒血流灌注成像评估狭窄效果,实验终点对小鼠股动脉进行取材,采用病理形态观察等方法判断股动脉斑块形成情况,组间比较采用非配对t检验。结果模型组小鼠股动脉(术前血流灌注-术后即刻血流灌注)/术前血流灌注高于对照组[(40.630±6.116)%比(3.000±1.852)%,t=16.650,P<0.01]。模型组小鼠股动脉可见明显的斑块形成且股动脉管腔狭窄严重,对照组小鼠股动脉未见明显斑块形成且股动脉管腔未见明显狭窄。模型组小鼠股动脉可获得管腔面积(内膜下面积)/内弹力板下面积低于对照组[(20.880±3.885)%比(67.890±3.530)%,t=20.030,P<0.01]。结论采用股动脉袖带置入联合串联结扎可有效构建股动脉粥样硬化疾病小鼠模型。Objective To construction an femoral artery atherosclerosis mouse model.Methods A total of 168-weeks-old male apolipoprotein E-deficient(ApoE-/-)mice purchased from Beijing Huafukang Biotechnology Corporation were randomly divided into two groups using a random number table:femoral artery cuff placement group(control group,n=8)and the femoral artery cuff placement combined with tandem stenosis group(model group,n=8).The mice in both groups were induced by a high-fat diet(HFD)for 4 weeks,then underwent surgical intervention separately,and then the HFD continued for another 8 weeks,for 12 weeks in the experimental cycle.Laser Doppler flow perfusion imaging was utilized to assess the stenosis effect,the femoral arteries of the mice were sampled at the end of the experiment,and the femoral artery plaque formation was judged by histopathology.The unpaired t-test was used for statistical methods.Results(Femoral artery preoperative perfusion-immediate postoperative perfusion)/preoperative perfusion was higher in the model group than in the control group[(40.630±6.116)%vs.(3.000±1.852)%,t=16.650,P<0.01].In contrast,there was no significant decrease in the control group.Pathomorphologic observations showed that the model group had apparent plaque formation and severe stenosis of the femoral artery lumen.In contrast,the control group had no apparent plaque formation or evident stenosis of the femoral artery lumen.The obtainable lumen area(area under the endothelium)/area under the internal elastic plate in the femoral artery of mice in the model group was lower than that in the control group[(20.880±3.885)%vs.(67.890±3.530)%,t=20.030,P<0.01].Conclusion Cuff placement combined with tandem stenosis can construct a mouse model of femoral atherosclerosis disease.
