摘 要:目的:探讨microRNA-374a(miR-374a)通过PTEN/AKT信号通路对乳腺癌细胞增殖与凋亡的调节作用。方法:采用实时荧光定量PCR(qRT-PCR)技术对人乳腺癌细胞MCF-7和正常乳腺细胞Hs 578Bst中的miR-374a和PTEN mRNA转录水平进行检测。此外,通过蛋白免疫印迹(Western blot)方法,测定MCF-7和Hs 578Bst细胞以及对照组和MiR-374a抑制剂处理组中PTEN和AKT及其磷酸化形式蛋白的表达变化。采用CCK-8法和TUNEL法,分别评估对照组和MiR-374a抑制剂处理组的细胞增殖和凋亡情况。同时,利用Western blot技术检测两组细胞中凋亡相关蛋白Bcl-2、Cleaved caspase-3和Cleaved caspase-9的表达变化。结果:在乳腺癌细胞中,MiR-374a的表达水平显著下调。通过转染技术抑制MiR-374a的表达后,乳腺癌细胞的增殖速率增快,凋亡率降低。同时,Western blot分析表明,抑制MiR-374a的表达导致PTEN蛋白水平下调,并伴随着AKT磷酸化水平的升高,从而激活PTEN/AKT信号通路。结论:MiR-374a通过靶向调节PTEN/AKT信号通路,对乳腺癌细胞的增殖和凋亡发挥重要的调节作用,为乳腺癌的治疗提供了新的潜在靶点和治疗策略。Objective:To investigate MicroRNA-374a(miR-374a)via PTEN/AKT signaling pathway to regulate breast cancer cell proliferation and apoptosis.methods:MiR-374a and PTEN mRNA transcript levels in human breast cancer cells MCF-7 and normal breast cells Hs 578Bst were examined by real-time fluorescence quantitative PCR(qRT-PCR).In addition,changes in the expression of PTEN and AKT and their phosphorylated forms of proteins in MCF-7 and Hs 578Bst cells as well as in control and MiR-374a inhibitor-treated groups were determined by protein immunoblotting(Western blot)Methods.Cell proliferation and apoptosis were assessed in control and MiR-374a inhibitor-treated groups using CCK-8 assay and TUNEL assay,respectively.Meanwhile,the expression changes of apoptosis-related proteins Bcl-2,Cleaved caspase-3 and Cleaved caspase-9 were detected in the cells of both groups adopting Western blot technique.Results:The expression level of MiR-374a was significantly downregulated in breast cancer cells.After inhibiting the expression of MiR-374a by transfection technique,the proliferation rate of breast cancer cells increased and the apoptosis rate decreased.Meanwhile,Western blot analysis showed that inhibition of MiR-374a expression led to the down-regulation of PTEN protein level,which was accompanied by an increase in the level of AKT phosphorylation,thus activating the PTEN/AKT signaling pathway.Conclusion:MiR-374a plays an important regulatory role in the proliferation and apoptosis of breast cancer cells by targeting and regulating the PTEN/AKT signaling pathway,which provides a new potential target and therapeutic strategy for the further treatment of breast cancer.
