摘 要:为了开发绿色且高效的木质纤维素处理新技术,以玉米秸秆为模型原料,将球磨预处理与蒸汽预处理相结合用以提高玉米秸秆水解效率。研究结果表明:球磨预处理可显著提高秸秆水解效率,在10%固载量条件下,4 mm秸秆经过球磨预处理后,水解后葡萄糖和还原糖的回收率分别由38%和36%提高至65%和58%;在球磨预处理的基础上,将秸秆浸入6 g·L-1NaOH并在121℃条件下蒸汽预处理1 h,水解后葡萄糖和还原糖的回收率均达到100%,且在此预处理条件下,当固载量由10%增加至30%时,葡萄糖和还原糖的回收率仍可达到77.14%和75.82%;另外,TOC和TN分析表明,在预处理条件优化和水解效率显著提高的同时,秸秆中的非糖副产物的水解率也显著提高,圆红酵母最高可在秸秆固载量为20%时获得的水解液中进行生长和油脂发酵,生物量、油脂产量和油脂含量分别为16.00 g·L-1,6.00 g·L-1和35.61%。To develop an efficient and environmentally friendly lignocellulosic pretreatment method,we utilized corn stover(CS)as a model material and proposed a combination of ball milling and steam pretreatments to enhance the CS digestibility.Our results demonstrated that the ball milling pretreatment of CS with 4 mm in diameter markedly improved its digestibility,increasing glucose and TRS recoveries from 38%and 36%to 65%and 58%at a solid loading of 10%,respectively.Additionally,100%recoveries of both glucose and TRS after enzymatic hydrolysis were achieved when the CS pretreated by the ball milling was further subjected to the steam pretreatment at 121°C for 1 hour after immersion in 6 g·L-1 NaOH.Even at a solid loading of 30%,the glucose and TRS recoveries remained high,reaching 77.14%and 75.82%,respectively,under these pretreatment conditions.However,TOC and TN analyses indicated a significant increase in the digestibility of non-sugar byproducts with the CS digestibility increasing,which had downstream impacts on the lipid fermentation process.Rhodosporidium toruloides could grow and produce lipid in the CS hydrolysate at a solid loading of 20%,with the final biomass,lipid and lipid content being 16.00 g·L-1,6.00 g·L-1 and 35.61%,respectively.
