摘 要:为建立油莎豆(Cyperus esculentus)快速繁殖体系,本试验以黑油莎2号为试验材料,茎尖作为外植体,采用植物不同组织培养条件培养油莎豆无菌苗,以期筛选出最佳茎尖消毒方法、适宜茎尖分化培养基配方以及快速生根培养基配方。结果表明:随消毒时间的增加,黑油莎2号茎尖污染率呈现降低的趋势,而存活率则呈现先升后降的变化;MS+0.9 mg/L 6-苄基腺膘呤(6-BA)+0.3 mg/L萘乙酸(NAA)处理时,黑油莎2号茎尖分化成苗数量显著高于其他处理(P <0.05),且茎尖分化成苗率最高,为62.67%;1/2MS+0.2 mg/L吲哚丁酸(IBA)处理时,黑油莎2号无菌苗生根数量(29.00)最高,显著高于其他处理(P <0.05),且根系粗壮,根长为0.9~3.0 cm。最后得出75%酒精30s+0.1%升汞4 min为黑油莎2号最佳茎尖消毒方法,MS+0.9 mg/L 6-BA+0.3 mg/L NAA为最佳茎尖分化培养基配方,MS+1.0 mg/L 6-BA和1/2MS+0.2 mg/L IBA分别是最佳快速繁殖和生根培养基配方。油莎豆快速繁殖体系构建为寒区高效繁育油莎豆优异种苗及种质资源创新提供依据。To construct the rapid propagation system for Cyperus esculentus,Heiyousha No.2 was taken the test material with its stem tip as explant to obtain the aseptic seedling of Cyperus esculentus through plant tissue culture technology,as well as screen the best sterilization method for stem tip,appropriate differential medium formula for stem tip and rapid rooting medium formula.According to the results,the contamination rate for stem tip of Heiyousha No.2 shows a reducing trend along with increasing sterilization time,and the survival rate showed a change from rising to descending;If carrying out MS+0.9 mg/L 6-benzyl adenine(6-BA)+0.3 mg/L 1-naphthaleneacetic acid(NAA)treatment,the seedling number differentiated from stem tip of Heiyousha No.2 was obviously higher than other treatment(P<0.05),and the seeding percentage differentiated from stem tip was the highest,which is 62.67%.If carrying out 1/2MS+0.2 mg/L Indole-3-butyric acid(IBA)treatment,the rooting number for aseptic seedling of Heiyousha No.2 was the highest(29.00),which was obviously higher than other treatment(P<0.05),and the root was thick and strong with the root length was 0.9~3.0 cm.In summary,75%ethyl alcohol 30 s+0.1%L HgCl24 min were the best sterilization method for stem tip of Heiyousha No.2,and MS+0.9 mg/L 6-BA+0.3 mg/L NAA was the best differential medium formula for stem tip,MS+1.0 mg/L 6-BA and 1/2MS+0.2 mg/L IBA were the best rapid propagation and rooting medium formulations,respectively.The construction of rapid propagation system for Cyperus esculentus provide a basis for the high-efficiency propagation and breeding of excellent seedings of Cyperus esculentus and germplasm resources innovation in cold regions.
