摘 要:目的探索枸杞籽油对亚急性衰老大鼠睾丸Nrf2/ARE通路和氧化损伤的影响。方法将60只8周龄♂SD大鼠中的50只(空白组10只)颈部皮下注射D-半乳糖125 mg·kg^(-1),持续8周建立亚急性衰老大鼠模型,β-半乳糖苷酶(β-galactosidase,β-gal)观察衰老细胞,HE观察睾丸形态,ELISA检测血清中睾酮(testosterone,T)、促卵泡激素(follicle stimulating hormone,FSH)和黄体生成素(luteinizing hormone,LH)及超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽(glutathione,GSH)和丙二醛(malondialdehyde,MDA)的水平,免疫组化和Western blot检测衰老、氧化损伤和Nrf2/ARE通路相关因子表达。结果模型鉴定成功后,经枸杞籽油干预,改善了睾丸形态,下调β-gal和γH2AX表达;升高血清中SOD、GSH以及T、FSH水平及降低MDA和LH水平(P<0.05);上调睾丸组织Nrf2、GCLC、NQO1和SOD2蛋白表达水平(P<0.05)以及支持细胞中Nrf2核表达。结论枸杞籽油激活亚急性衰老大鼠睾丸Nrf2/ARE信号通路表达,降低其氧化损伤,改善其激素水平。Aim To explore the effects of Lycium berry seed oil on Nrf2/ARE pathway and oxidative damage in testis of subacute aging rats.Methods Fifty out of 60 male SD rats,aged 8 weeks,were subcutaneously injected with 125 mg·kg^(-1) D-galactosidase in the neck for 8 weeks to establish a subacute senescent rat model.The presence of senescent cells was observed usingβ-galactosidase(β-gal),while testicular morphology was examined using HE staining.Serum levels of testosterone(testosterone,T),follicle-stimulating hormone(follicle stimulating hormone,FSH),luteinizing hormone(luteinizing hormone,LH),superoxide dismutase(superoxide dismutase,SOD),glutathione(glutathione,GSH)and malondialdehyde(malondialdehyde,MDA)were measured through ELISA,and the expressions of factors related to aging,oxidative damage,and the Nrf2/ARE pathway were assessed via immunohistochemical analysis and Western blotting.Results After successfully identifying the model,the morphology of the testis was improved and the intervention of Lycium seed oil led to a down-regulation in the expression ofβ-gal andγH2AX.The serum levels of SOD,GSH,T,and FSH increased while MDA and LH decreased(P<0.05).Additionally,there was an up-regulated expression of Nrf2,GCLC,NQO1,and SOD2 proteins in testicular tissue(P<0.05)and nuclear expression of Nrf2 in sertoli cells.Conclusion Lycium barbarum seed oil may reduce oxidative damage in testes of subacute senescent rats by activating the Nrf2/ARE signaling pathway.
