摘 要:目的:探讨乳腺癌(breast cancer,BC)患者癌组织中转录因子BTB-CNC同源体1(BTB and CNC homology,BACH 1)、血红素环氧化酶1(hemoxygenas-1,HO-1)的表达及与乳腺癌临床特征的相关性,进一步研究丹皮酚(paeonol,Pae)对BACH 1、HO-1的表达调控。方法:免疫组织化学方法检测乳腺癌组织与癌旁组织中BACH 1、HO-1分子的表达,分析BACH 1、HO-1表达与患者临床病理因素之间的关系;体外培养人MDA-MB-468乳腺癌细胞,给予丹皮酚处理48 h,通过CCK-8、Transwell检测细胞的增殖、迁移,Western blot检测BACH 1、HO-1的表达。结果:与非癌组织相比,BACH 1在乳腺癌组织中低表达,与TNM分级显著相关(P<0.05)。HO-1高表达与肿瘤分级显著相关(P<0.05)。CCK-8、Transwell结果显示丹皮酚抑制细胞增殖并减少肿瘤细胞迁移。Western blot结果显示丹皮酚60 mg/L使BACH 1表达上调,HO-1表达下降。结论:BACH 1、HO-1参与乳腺癌发病过程,可作为乳腺癌的预后参考标志物。丹皮酚可抑制乳腺癌细胞增殖迁移,可能与调控BACH 1、HO-1的表达有关。Objective:To investigate the expression of transcription factors BTB and CNC homology 1(BTB and CNC homology,BACH 1)and hemoxygenas-1(HO-1)in Cancer tissues of patients with Breast Cancer(BC)and their correlation with clinical features of breast cancer.To further study the regulation of Paeonol(Pae)on the expression of BACH 1 and HO-1.Methods:The expression of BACH 1 and HO-1 molecules in breast cancer tissues and adjacent tissues was detected by immunohistochemical method,and the relationship between the expression of BACH 1 and HO-1 and clinical pathological factors of patients was analyzed.Human MDA-MB-468 breast cancer cells were cultured in vitro and treated with paeonol for 48 h.The proliferation and migration of the cells were detected by CCK-8 and Transwell,and the expressions of BACH 1 and HO-1 were detected by Western blot.Results:Compared with non-cancer tissues,the expression of BACH 1 was lower in breast cancer tissues and significantly correlated with TNM grade(P<0.05).The high expression of HO-1 was significantly correlated with tumor grade(P<0.05).CCK-8 and Transwell results showed that paeonol inhibited cell proliferation and reduced tumor cell migration.Western blot results showed that paeonol 60 mg/L increased the expression of BACH 1 and decreased the expression of HO-1.Conclusion:BACH 1 and HO-1 are involved in the pathogenesis of breast cancer and can be used as prognostic reference markers for breast cancer.Paeonol can inhibit the proliferation and migration of breast cancer cells,which may be related to the regulation of the expression of BACH 1 and HO-1.
