摘  要:目的:探讨结肠癌患者癌组织中淋巴增强子结合因子1(lymphatic enhancer binding factor 1,LEF1)的表达水平及其临床意义。方法:免疫组化染色法结合Western blot观察结肠癌患者癌组织和癌旁组织中LEF1的表达情况,并对癌组织中LEF1的表达与结肠癌患者临床病理特征的相关性进行统计学分析。Kaplan-Meier生存分析LEF1的表达与结肠癌患者预后的关联,多因素COX回归分析影响结肠癌患者生存预后的危险因素。进一步分析TCGA数据库结肠癌患者癌和癌旁组织中LEF1的表达水平,及其表达与患者预后以及淋巴结转移的相关性。此外,细胞水平采用Western blot检测结肠癌细胞系中LEF1的蛋白表达情况,并通过转染LEF1 siRNA观察对结肠癌HCT116细胞生物学功能的影响。结果:结肠癌组织中LEF1的表达较癌旁组织显著增加(P<0.05),且其表达水平与肿瘤病理分期、分化程度和淋巴结转移呈正相关(P<0.05),与患者年龄、病理类型无关(P>0.05)。LEF1高表达结肠癌患者的生存期明显低于低表达患者(P<0.05)。多因素COX回归分析结果显示,LEF1高表达、肿瘤病理分期、肿瘤分化程度以及淋巴结转移是结肠癌患者不良预后的危险因素(P<0.05)。TCGA数据库分析结果显示,结肠癌组织中LEF1的表达水平显著高于癌旁组织(P<0.05),且其表达水平与患者预后呈负相关(P<0.05),而与患者淋巴结转移呈正相关(P<0.05)。此外,细胞实验结果显示,人结肠癌细胞系中LEF1的表达水平显著高于正常结肠上皮细胞(P<0.05),敲减LEF1可有效降低结肠癌HCT116细胞的迁移和侵袭能力,并抑制Wnt/β-catenin信号通路活性(P<0.05)。结论:LEF1高表达与结肠癌患者的短生存期和不良预后密切相关,可作为结肠癌患者术后复发和转移以及临床预后的重要生物标志物。Objective:To investigate the expression level and clinical significance of lymphatic enhancer binding factor 1(LEF1) in colon cancer patients.Methods:Expression level of LEF1 in tumor and adjacent tissues of colon cancer patients was observed by immunohistochemical staining combined with Western blot,and the correlation between LEF1 expression in tumor tissues and the clinicopathological characteristics of colon cancer patients was statistically analyzed.Kaplan-Meier survival analysis was used to analyze the effect of LEF1 expression level on the prognosis of colon cancer patients,and multivariate COX regression was used to analyze the risk factors affecting the prognosis of colon cancer patients.The expression level of LEF1 in cancer and adjacent tissues of colon cancer patients in the TCGA database was further analyzed,and the correlation between LEF1 expression and patient prognosis and lymphatic metastasis was analyzed.In addition,at the cellular level,Western blot was used to detect LEF1 protein expression in colon cancer cell lines,and the effect of LEF1 siRNA transfection on the biological function of colon cancer HCT116 cells was observed.Results:LEF1 expression level in tumor tissues of colon cancer patients was significantly increased compared with that in paracancer tissues(P<0.05),and its expression level was positively correlated with tumor pathological stage,degree of differentiation,and lymphatic metastasis(P<0.05),but not with patient age,pathological type(P>0.05).In addition,the survival time of colon cancer patients with high LEF1 expression levels was significantly lower than that of patients with low LEF1 expression levels( P < 0.05).Multivariate COX regression analysis showed that high LEF1 expression level,tumor pathological stage,degree of tumor differentiation,and lymph node metastasis were risk factors for poor prognosis of colon cancer patients( P < 0.05).The results of TCGA database analysis showed that the expression level of LEF1 in colon cancer tissues was significantly higher than t
