摘  要:目的探究白细胞介素11对急性肝衰竭大鼠肝细胞凋亡及JAK/STAT信号通路的影响。方法将40只大鼠随机分为NC组、ALF组、IL-11组和IL-11+colivelin组。ELISA检测肝功能和炎症水平;HE染色检测肝组织病理学变化;TUNEL检测肝细胞凋亡指数;蛋白质印迹法检测肝组织中JAK2、p-JAK2、STAT3、p-STAT3蛋白表达;qRT-PCR检测肝组织中JAK2、STAT3 mRNA相对表达量。结果和NC组相比,ALF组大鼠血清中ALT、AST、TBIL含量、TNF-α、IL-1β、IL-6水平、肝细胞凋亡指数、肝组织中p-JAK2/JAK2和p-STAT3/STAT3比值及JAK2、STAT3 mRNA相对表达量均明显升高(P<0.05);和ALF组相比,IL-11组大鼠血清中ALT、AST、TBIL含量、TNF-α、IL-1β、IL-6水平、肝细胞凋亡指数、肝组织中p-JAK2/JAK2和p-STAT3/STAT3比值及JAK2、STAT3 mRNA相对表达量均明显降低(P<0.05);和IL-11组相比,IL-11+colivelin组大鼠血清中ALT、AST、TBIL含量、TNF-α、IL-1β、IL-6水平、肝细胞凋亡指数、肝组织中p-JAK2/JAK2和p-STAT3/STAT3比值及JAK2、STAT3 mRNA相对表达量均明显升高(P<0.05)。结论IL-11可能通过抑制JAK2/STAT3通路激活,抑制炎症反应和肝细胞凋亡,从而改善大鼠急性肝衰竭。Objective To investigate the effects of interleukin 11 on hepatocyte apoptosis and JAK/STAT signaling pathway in rats with acute liver failure.Methods Forty rats were randomly divided into NC group,ALF group,IL-11 group,and IL-11+colin group.ELISA detection of liver function and inflammation levels;HE staining was used to detect pathological changes in liver tissue;TUNEL detection of liver cell apoptosis index;Protein blotting was used to detect the expression of JAK2,p-JAK2,STAT3,and p-STAT3 proteins in liver tissue;QRT PCR was used to detect the relative expression levels of JAK2 and STAT3 mRNA in liver tissue.Results Compared with the NC group,the serum levels of ALT(365.71±40.62),AST(514.87±53.41)and TBIL(30.95±4.06),the levels of TNF-α(314.65±35.19),IL-1β(296.17±30.84)and IL-6(118.42±12.31),the apoptotic index of hepatocytes(13.28±1.46),the p-JAK2/JAK2(0.89±0.11)and p-STAT3/STAT3(0.95±0.12)ratios,and the relative expression of JAK2(1.94±0.21)and STAT3(1.89±0.20)mRNA in liver tissues of rats in the ALF group were all significantly elevated(P<0.05);Compared with the ALF group,the serum levels of ALT(94.18±10.35),AST(194.21±20.83)and TBIL(3.11±0.48),the levels of TNF-α(98.47±10.63),IL-1β(91.62±10.17)and IL-6(36.17±4.08),the apoptotic index of hepatocytes(2.11±0.32),the p-JAK2/JAK2(0.34±0.05)and p-STAT3/STAT3(0.46±0.06)ratios,and the relative expression of JAK2(1.31±0.15)and STAT3(1.26±0.14)mRNAs in the liver tissues of the rats in the IL-11 group were significantly lower(P<0.05);Compared with the IL-11 group,the serum levels of ALT(321.48±34.51),AST(468.32±50.17)and TBIL(27.94±3.11),the levels of TNF-α(282.31±30.45),IL-1β(247.62±25.17)and IL-6(102.43±11.41),the apoptotic index of hepatocytes(12.81±1.37),the p-JAK2/JAK2(0.80±0.10)and p-STAT3/STAT3(0.89±0.10)ratios,and the relative expression of JAK2(1.89±0.19)and STAT3(1.80±0.19)mRNAs in the liver tissues of the rats in the IL-11+colivelin group were significantly elevated.(P<0.05).Conclusion IL-11 may ameliorate acute live
