摘  要:Rho激酶(ROCK)是Rho家族的关键效应器蛋白,通过Rho/ROCK信号通路在多种细胞生物学过程中发挥关键作用,包括细胞形态改变、细胞骨架重组、细胞运动以及细胞-细胞和细胞-基质相互作用等。Rho/ROCK信号通路的异常激活可介导多种疾病的发生、发展,并与这些疾病的病理过程相关。ROCK抑制剂通过抑制Rho/ROCK信号通路发挥神经保护、抗炎、抗肿瘤和抗纤维化等作用,因此其在多种疾病中具有潜在治疗价值。ROCK抑制剂在眼部疾病(青光眼、角膜疾病和视网膜疾病等)治疗中展现出巨大潜力,或可为这些疾病的治疗提供一种全新的策略和方法。Rho kinase(ROCK)is a key effector protein in the Rho family,playing a crucial role in various cellular biological processes through the Rho/ROCK signaling pathway,including cellular morphological changes,cytoskeletal reorganization,cell motility,and cell-cell and cell-matrix interactions.Abnormal activation of the Rho/ROCK signaling pathway can mediate the occurrence and development of various diseases,and is associated with the pathological processes of these diseases.ROCK inhibitors exert neuroprotective,anti-inflammatory,anti-tumor,and anti-fibrotic effects by inhibiting the Rho/ROCK signaling pathway,thus having potential therapeutic value in various diseases.In the treatment of eye diseases such as glaucoma,corneal diseases and retinal diseases,ROCK inhibitors have shown great potential and may provide a new strategy and method for the treatment of these diseases.
