摘  要:目的探究新藤黄酸对急性淋巴细胞性白血病细胞Nalm-6的增殖、迁移、侵袭的影响及作用机制。方法使用不同浓度的新藤黄酸处理Nalm-6细胞,通过MTT实验、软琼脂克隆形成实验、transwell迁移实验与transwell侵袭实验来检测新藤黄酸对Nalm-6细胞增殖、迁移、侵袭的影响。通过凋亡坏死检测试剂盒检测Nalm-6细胞的凋亡情况,通过Western blot实验检测Nalm-6细胞中c-Myc、P21蛋白表达情况。结果新藤黄酸可呈剂量依赖性抑制Nalm-6细胞的增殖、克隆形成、迁移与侵袭,导致Nalm-6细胞凋亡,下调c-Myc表达,上调P21表达。结论新藤黄酸可通过调控c-Myc和P21抑制Nalm-6,有望为急性淋巴细胞性白血病提供新的潜在治疗选择。Objective To investigate the effects and mechanisms of neogambogic acid on proliferation,migration and invasion of acute lymphoblastic leukemia cells Nalm-6.Methods Nalm-6 cells were treated with neogambogic acid at different concentrations.MTT assay,soft agar colony formation assay,transwell migration assay and transwell invasion assay were used to evaluate the effects of neogambogic acid on proliferation,migration and invasion of Nalm-6 cells.Apoptosis and necrosis assay kit was conducted to detect the apoptosis of Nalm-6 cells,and western blot was used to determine the protein expression of c-Myc and P21 in Nalm-6 cells.Results Neogambogic acid inhibited proliferation,colony formation,migration and invasion of Nalm-6 cells in a dose-dependent manner,and induced the apoptosis of Nalm-6 cells,down-regulated the expression of c-Myc and up-regulated the expression of P21.Conclusion Neogambogic acid can regulate c-Myc and P21 to inhibit Nalm-6 cells,which may provide a new therapeutic strategy for acute lymphoblastic leukemia.
