恢复正常饮食对高脂高胆固醇饮食诱导小鼠脂肪肝的改善作用

摘  要:目的 观察恢复正常饮食对高脂高胆固醇(HFHC)饮食诱导非酒精性脂肪性肝病(NAFLD)的改善作用,并探讨其中的作用机制。方法 HFHC饮食16w诱导建立小鼠NAFLD模型后随机分为模型对照组和恢复正常饮食干预组,并设正常对照组。模型对照组饲喂HFHC饲料,恢复正常饮食干预组饲喂正常饲料,正常对照组饲喂正常饲料。10 w后处死所有小鼠,检测小鼠血脂水平、血清胆汁酸组成、肝功能和组织形态学差异。16S rDNA测序分析盲肠内容物中肠道菌群的组成。结果 模型对照组小鼠的血清总胆固醇、丙氨酸氨基转移酶、天冬氨酸氨基转移酶和低密度脂蛋白胆固醇水平较正常对照组小鼠显着升高,恢复正常饮食干预显着降低上述指标(P<0.05)。在模型对照组小鼠肝组织可见明显脂肪变性和气球样变,而恢复正常饮食干预可有效改善NAFLD活动性评分(P<0.05)。此外,恢复正常饮食干预有助于降低NAFLD小鼠血清胆汁酸疏水指数,减少盲肠内容物中Ruminococcaceae_UCG-010丰度,增加Anaerotruncus和Ruminococcaceae_NK4A214_group丰度(P<0.05)。结论 恢复正常饮食可通过影响胆汁酸代谢和肠道菌群组成改善HFHC饮食诱导的NAFLD。Objective To observe the protective effect on high-fat and high-cholesterol(HFHC) diet induced non-alcoholic fatty liver disease(NAFLD) in mice by switching back to normal diet feeding and explore the mechanism.Methods Mice were fed a HFHC diet for 16 weeks to establish a NAFLD model and then randomly divided into two groups:HFHC+NC group switched back to normal diet feeding;HFHC group continued HFHC diet feeding.The control group was continuously fed a normal chow diet.After 10 weeks,all mice were sacrificed and serum lipids,the composition of serum bile acids,liver function and histomorphology were determined.The composition of gut microbiota in the cecum contents were profiled using 16S rDNA gene sequencing.Results Compared with the control group,serum levels of total cholesterol,alanine transaminase,aspartate transaminase and low-density lipoprotein cholesterol in the HFHC group were significantly increased.Switching back to normal diet feeding significantly reduced these indicators as compared to the HFHC group(P<0.05).The HFHC group had obvious hepatic steatosis and balloon degeneration when compared with the control group,whereas switching back to normal diet feeding alleviated the NAFLD activity score(P<0.05).In addition,switching back to normal diet feeding could decrease hydrophobicity index of serum bile acids and the abundance of Ruminococcaceae UCG-_010 of cecum contents,and increase the abundance of Anaerotruncus and Ruminococcaceae_NK4A214_group of cecum contents(P<0.05).Conclusion Switching back to normal diet feeding can alleviate HFHC diet-induced NAFLD via regulating gut microbiota and serum bile acids composition.
