摘 要:目的 探讨缬草酸对口腔癌KB细胞恶性生物学行为的影响及其相关机制。方法 将口腔癌KB细胞分为5组,分别用0、1、2.5、5、10μmol/L的缬草酸处理细胞,Transwell实验分析不同实验组细胞的侵袭能力,细胞划痕实验分析不同实验组细胞迁移能力,TUNEL染色检测不同实验组细胞凋亡比例,蛋白质免疫印迹分析上皮-间充质转化(EMT)标记物表达情况以及Notch信号通路相关蛋白表达。结果 缬草酸能够显著降低口腔癌KB细胞的集落形成、癌细胞活性和促进细胞凋亡,抑制KB细胞的转移和侵袭能力;上调KB细胞E-cadherin的表达,抑制N-cadherin、Snail和Slug的表达;抑制口腔癌KB细胞Notch1、 Notch2、 Jagged1、 Hes1、 Nicastrin和Dll4的表达。结论 缬草酸通过调控Notch信号通路抑制口腔癌KB细胞的活性、转移以及侵袭能力。Objective To investigate the effect and related mechanism of valeric acid on the malignant biological behavior of oral cancer KB cells.Methods Oral cancer KB cells were divided into 5 groups,and the cells were treated with 0,1,2.5,5 and 10 mol/L valeric acid respectively.The invasion ability of cells in different experimental groups was analyzed by Transwell assay.The migration ability of cells in different experimental groups was analyzed by cell scratch assay.The apoptosis ratio of cells in different experimental groups was detected by TUNEL staining.The expressions of epithelial-mesenchymal transition(EMT)markers and the expressions of Notch signaling pathway-related proteins were analyzed by Western blot.Results Valerenic acid could significantly reduce the colony formation,cell viability and apoptosis of KB cells,inhibit the metastasis and invasion of KB cells,up-regulate the expressions of E-cadherin,and inhibit the expressions of N-cadherin,Snail and Slug in KB cells,inhibit the expressions of Notch1,Notch2,Jagged1,Hes1,Nicastrin and Dll4 in oral cancer KB cells.Conclusion Valeric acid can inhibit the activity,metastasis and invasion of oral cancer KB cells by regulating Notch signaling pathway.
