摘 要:目的 探讨金银花多糖对三阴性乳腺癌细胞增殖、凋亡、迁移的影响。方法 三阴性人乳腺癌细胞株MDA-MB-231,设为空白对照组和金银花多糖干预A组、B组、C组、D组。空白对照组不作处理,金银花多糖干预各组分别加入浓度为200 mg/mL、400 mg/mL、600 mg/mL、800 mg/mL的金银花多糖20μL。24h、48h后,应用MTT实验检测细胞增殖情况、划痕实验检测细胞迁移情况、TUNEL法检测细胞凋亡情况、RT-PCR法检测Bcl-2、Bax mRNA表达情况。结果 金银花多糖干预组的增殖率、迁移距离、Bcl-2 mRNA的表达水平均明显低于空白对照组(P<0.05);凋亡率、Bax mRNA表达水平明显高于空白对照组(P<0.05)。金银花多糖浓度越高,增殖率、迁移距离、Bcl-2 mRNA的表达水平越低,凋亡率、Bax mRNA的表达水平越高(P<0.05)。金银花多糖干预组48 h时的增殖率、Bcl-2 mRNA的表达水平明显低于24 h;凋亡率、Bax mRNA的表达水平明显高于24h(P<0.05)。结论 金银花多糖对三阴性乳腺癌细胞的增殖与迁移具有明显的抑制作用,对其凋亡具有明显的促进作用,其作用机制与对Bcl-2/Bax凋亡途径的调节密切相关。Objective To investigate the effects of Honeysuckle polysaccharide on the proliferation,apoptosis and migration of triple-negative breast cancer cells.Methods The triple-negative human breast cancer cell line MDA-MB-231 was divided into a blank control group and four intervention groups(A,B,C,D)treated with different concentrations of Honeysuckle polysaccharide(200 mg/mL,400 mg/mL,600 mg/mL,800 mg/mL).The control group received no treatment,and the intervention groups were treated with 20μL of the respective concentrations of Honeysuckle polysaccharide.After 24 hours and 48 hours,the cell proliferation was detected using MTT assay,cell migration was detected using a scratch assay,apoptosis was detected using TUNEL staining,and the expression levels of Bcl-2 and Bax mRNA were detected using RT-PCR.Results The proliferation rate,migration distance,and expression level of Bcl-2 mRNA in the intervention groups were significantly lower than those in the control group(P<0.05).The apoptosis rate and expression level of Bax mRNA in the intervention groups were significantly higher than those in the control group(P<0.05).With increasing concentration ofHoneysuckle polysaccharide,the proliferation rate,migration distance and expression level of Bcl-2 mRNA decreased,while the apoptosis rate and expression level of Bax mRNA increased(P<0.05).The proliferation rate and expression level of Bcl-2 mRNA in the intervention groups were significantly lower at 48 hours than at 24 hours(P<0.05).The apoptosis rate and expression level of Bax mRNA were significantly higher at 48 hours than at 24 hours(P<0.05).Conclusion Honeysuckle polysaccharide inhibits the proliferation and migration of triple-negative breast cancer cells and promotes their apoptosis.This effect is closely related to the regulation of Bcl-2/Bax apoptotic pathway.
