摘  要:目的通过对2021—2022年北京市通州区农贸市场的外环境及相关人员标本的禽流感病毒检测结果及基因特征分析,为我市禽流感防控策略提供依据。方法2021—2022年共采集外环境及食品涂抹标本2184例,相关人员咽拭子标本124例。利用实时荧光RT-PCR法对标本进行核酸检测,采用illumina二代测序平台对病毒载量高的阳性标本进行全基因组测序,分析其基因特征。结果实时荧光RT-PCR核酸检测结果显示,外环境及食品涂抹标本检出2例H5N8阳性、78例H7N9阳性、177例H9N2阳性、2例H5N8和H9N2共感染、1例H7N9和H9N2共感染,124例人员标本检测结果均为阴性。对病毒载量高的阳性标本进行全基因组测序分析,1例H5N8病毒的HA基因属于为2.3.4.4分支,HA基因的HA0裂解位点序列为多个连续性碱性氨基酸(REKRRKR/GLF),2例H7N9病毒的HA基因与Yangtza river delta属于同一分支,HA基因的HA0裂解位点序列为多个连续性碱性氨基酸(VPKRKRTAR/GLF),均属于高致病性禽流感病毒。遗传进化树分析提示,H5N8和H7N9的HA基因与国内报道的病毒株亲缘关系较近。结论农贸市场是禽流感病毒分布的主要场所,高致病性及低致病性禽流感病毒同时空交叠感染现状,提示应加强禽流感病毒的监测防控工作及防范知识的宣传。Objective To analyze the test results and genetic characteristics of avian influenza virus in the samples collected from environment and related staff in a farmer’s market in Tongzhou district of Beijing from 2021 to 2022,so as to provide basis for the prevention and control strategy of avian influenza in our city.Methods From 2021 to 2022,a total of 2184 swab samples were collected from environment and food and 124 throat swab samples from relevant staff.Real-time RT-PCR was used to detect viral nucleic acid in samples.The next generation sequencing platform from illumina was used in whole genome sequencing for the positive samples with high virus titer and the genetic characteristics were analyzed.Results The results detected by real-time RT-PCR showed that among samples from environment and food,2 were positive for H5N8,78 were positive for H7N9,177 were positive for H9N2,2 were co-infected with H5N8 and H9N2 and 1 was co-infected with H7N9 and H9N2.All 124 human samples were negative.The genome sequence analysisof high viral titer samples were performed.The HA gene in one case of H5N8 virus belonged to the 2.3.4.4 clade,and the HA0 cleavage site sequence of the gene consisted of multiple continuous basic amino acids(REKRRKR/GLF).The HA gene in 2 cases of H7N9 virus belonged to the same branch as Yangtza river delta,and the HA0 cleavage site sequence of the HA gene consists of multiple continuous basic amino acids(VPKRKRTAR/GLF),which belonged to highly pathogenic avian influenza viruses.The results of phylogenetic analysis showed that the HA genes of H5N8 and H7N9 were closely related to the virus strains reported in China.Conclusions The farmer’s market was the main place for the distribution of avian influenza virus.Simultaneous and spatial overlapping infection status of highly pathogenic and low pathogenic avian influenza viruses suggested that surveillance and control of avian influenza virus and propaganda of prevention knowledge should be strengthened.
