摘 要:目的 APOBEC3B的异常激活与多种癌症的发生发展密切相关,本研究利用生物信息学技术分析APOBEC3B在肺鳞癌中的表达情况及功能。方法 利用TCGA比较不同分组间APOBEC3B的表达水平。通过Kaplan-Meier评估APOBEC3B表达对肺鳞癌预后的意义。使用GSEA对APOBEC3B高、低表达组样本进行功能富集分析。使用“CIBERSORT”评估分析组间免疫细胞浸润比例的差异。分析组间免疫相关功能的差异。分析组间TIDE评分的差异。计算APOB3C3B高、低表达组基因突变情况,分析肿瘤突变负荷(TMB)的差异。结果 与正常组织相比,肺鳞癌组织中APOBEC3B的表达明显上调。其表达水平与年龄、性别、临床分期、TNM分期均无显著相关性,UALCAN数据库的分析结果显示吸烟患者组、TP53突变组中APOBEC3B的表达显著上调。Kaplan-Meier生存分析显示,APOBEC3B高、低表达组间生存时间无显著差异。GSEA结果显示,APOBEC3B高表达组主要富集在DNA复制等过程。免疫相关功能的分析结果表明,APOBEC3B高、低表达组间在APC共抑制和嵌合共刺激受体功能存在差异。CIBERSORT分析显示,仅调节T细胞和中性粒细胞的浸润比例在APOBEC3B高、低表达组间存在差异。TIDE评分显示,APOBEC3B低表达组的TIDE评分显著高于高表达组,高、低表达组之间的TMB差异不显著。结论 APOBEC3B在肺鳞癌组织中高表达,与TP53基因突变相关,促进肿瘤的发生发展。APOBEC3B可作为肺鳞癌预后判断的生物标志物。Objective The abnormal activation of APOBEC3B is closely related to the occurrence and development of a variety of cancers.In this study,the expression and function of APOBEC3B in lung squamous cell carcinoma were analyzed by bioinformatics technology.Methods TCGA was used to compare the expression level of APOBEC3B in different groups.Kaplan-Meier was used to evaluate the prognostic significance of APOBEC3B expression in lung squamous cell carcinoma.GSEA was used to analyze the functional enrichment of APOBEC3B high and low expression groups.CIBERSORT was used to assess differences in the proportion of immune cells infiltrating between the groups.Differences in immune-related functions between groups were analyzed.Differences in TIDE scores between groups were analyzed.The mutations of APOB3C3B high and low expression groups were calculated,and the difference of tumor mutation burden(TMB)was analyzed.Results The expression of APOBEC3B in lung squamous cell carcinoma tissues was significantly up-regulated compared with that in normal tissues.There was no significant correlation between APOBEC3B expression and age,gender,clinical stage and TNM stage.UALCAN database analysis showed that APOBEC3B expression was significantly up-regulated in the smoking group and TP53 mutation group.Kaplan-Meier survival analysis showed that there was no significant difference in survival time between high and low APOBEC3B expression groups.GSEA results showed that APOBEC3B high expression group was mainly enriched in DNA replication and other processes.The results of immune-related function analysis showed that there were differences in APC coinhibition and chimeric costimulatory receptor function between high and low APOBEC3B expression groups.CIBERSORT analysis showed that only the percentage of infiltration of regulated T cells and neutrophils differed between the high and low APOBEC3B expression groups.TIDE score of APOBEC3B low expression group was significantly higher than that of APOBEC3B high expression group.There was no si
