摘 要:目的研究骨肉瘤(osteosarcoma)患者癌组织中热休克蛋白90 ATP酶激活因子1(the activator of HSP90 ATPase-1,AHA1)和赖氨酰氧化酶样蛋白2(lysyl oxidase like-2 protein,LOXL2)表达与侵袭转移基因mRNA表达的相关性及临床意义。方法选取2016年2月~2017年3月华北医疗健康集团峰峰总医院诊治的90例骨肉瘤患者为研究对象。应用实时荧光定量PCR检测组织中AHA1 mRNA,LOXL2 mRNA及侵袭转移基因Wnt家族成员9A(Wnt9a)mRNA,锌指E盒结合同源盒1(Zinc finger E-box binding homeobox 1,ZEB1)mRNA,锌指E盒结合同源盒2(ZEB2)mRNA,N-钙黏素(N-cad)mRNA和波形蛋白(Vim)mRNA表达。采用Pearson相关分析,比较不同临床特征骨肉瘤患者AHA1 mRNA,LOXL2 mRNA表达差异。Kaplan-Meier生存分析影响AHA1 mRNA,LOXL2 mRNA表达对骨肉瘤患者预后的影响。单因素及多因素COX回归分析影响骨肉瘤患者预后的因素。结果骨肉瘤组织中AHA1 mRNA(3.16±0.59),LOXL2 mRNA(2.84±0.44)及侵袭转移基因Wnt9a mRNA(3.23±0.42),ZEB1 mRNA(2.73±0.39),ZEB2 mRNA(2.52±0.56),N-cad mRNA(2.71±0.65),Vim mRNA(2.81±0.73)表达均高于癌旁组织(1.10±0.21,0.95±0.18,0.79±0.15,0.64±0.11,0.98±0.19,0.68±0.14,0.72±0.15),差异具有统计学意义(t=31.206,37.716,51.903,48.931,24.706,28.964,26.605,均P<0.05)。骨肉瘤组织中AHA1 mRNA与LOXL2 mRNA表达呈显著正相关(r=0.712,P<0.05)。骨肉瘤组织中AHA1 mRNA,LOXL2 mRNA与侵袭转移基因Wnt9a mRNA,ZEB1 mRNA,ZEB2 mRNA,N-cad mRNA,Vim mRNA表达呈显著正相关(r=0.504~0.720,均P<0.05)。Eneeking分期Ⅲ期、有软组织浸润和有肺转移骨肉瘤组织中AHA1 mRNA,LOXL2 mRNA表达高于Eneeking分期Ⅰ~Ⅱ期、无软组织浸润和无肺转移患者,差异具有统计学意义(t=14.122~171.054,均P<0.05)。AHA1 mRNA高表达组和低表达组患者五年生存率分别为36.36%(16/44),78.26%(36/46)。AHA1 mRNA高表达组患者五年累积生存率明显低于低表达组,差异有统计学意义(Log-rankχ^(2)=16.081,P<0.05)。LOXL2 mRNA高表达组Objective To investigate the correlation between the expression of the activator of HSP90 ATPase-1(AHA1),lysyl oxidase like-2 protein(LOXL2)in osteosarcoma tissues with mRNA expression of invasion and metastasis genes and their clinical significance.Methods A total of 90 osteosarcoma patients diagnosed and treated in North China Medical and Health Group Fengfeng General Hospital from February 2016 to March 2017 were selected as the research object.The expression of AHA1 mRNA,LOXL2 mRNA and invasion and metastasis genes Wnt family member 9A(Wnt9a)mRNA,zinc finger E-box binding homologous box 1(ZEB1)mRNA,zinc finger E-box binding homologous box 1(ZEB2)mRNA,N-cadherin(N-cad)mRNA,and vimentin(Vim)mRNA in tissues were detected by real-time fluorescence quantitative PCR.Pearson correlation analysis was used for correlation analysis.The differences in expression of AHA1 mRNA and LOXL2 mRNA in osteosarcoma patients among different clinical characteristics were compared.Kaplan-Meier survival analysis was used to analyze the effect of AHA1 mRNA and LOXL2 mRNA on the prognosis of osteosarcoma patients.The prognostic factors of osteosarcoma patients were analyzed by univariate and multivariate COX regression.Results The expressions of AHA1 mRNA(3.16±0.59),LOXL2 mRNA(2.84±0.44)and invasion and metastasis genes[Wnt9a mRNA(3.23±0.42),ZEB1 mRNA(2.73±0.39),ZEB2 mRNA(2.52±0.56),N-cad mRNA(2.71±0.65)and Vim mRNA(2.81±0.73)]in osteosarcoma tissues were higher than those in paracancerous tissues(1.10±0.21,0.95±0.18,0.79±0.15,0.64±0.11,0.98±0.19,0.68±0.14,0.72±0.15),and the differences were statissically significant(t=31.206,37.716,51.903,48.931,24.706,28.964,26.605,all P<0.05).There was a significant positive correlation between AHA1 mRNA and LOXL2 mRNA expression in osteosarcoma(r=0.712,P<0.05).The expressions of AHA1 mRNA and LOXL2 mRNA were significantly positively correlated with the expressions of invasion and metastasis genes(Wnt9a,ZEB1,ZEB2,N-cad,and Vim mRNA)in tumor tissue of osteosarcoma group(r=0.504~0.720,al
