摘 要:目的:研究超声微泡介导miR-545-3p通过调节胞裂蛋白2(septin 2,SEPT2)抑制乳腺癌恶性进展的机制。方法:以实时荧光定量PCR和免疫印迹实验检测人正常乳腺上皮细胞MCF-10A、人乳腺癌细胞MCF-7、HS-578T及HCC-1937细胞中miR-545-3p、SEPT2表达。体外培养MCF-7细胞并随机分为对照组、空微泡组、miR-545-3p微泡组、miR-545-3p+SEPT2过表达微泡组,分组处理后以实时荧光定量PCR和免疫印迹实验检测各组细胞miR-545-3p、SEPT2表达;以CCK-8、集落形成实验与流式细胞实验检测各组细胞增殖与凋亡;以细胞划痕、Transwell侵袭实验检测各组细胞迁移与侵袭。于裸鼠腹壁皮下接种MCF-7细胞构建其移植瘤模型并以同样方法分组,经分组处理后检测各组裸鼠肿瘤体积与质量。结果:与MCF-10A细胞相比,MCF-7、HS-578T及HCC-1937细胞中miR-545-3p表达降低(P<0.05),SEPT2 mRNA与蛋白表达升高(P<0.05)。与对照组相比,miR-545-3p微泡组细胞miR-545-3p表达、凋亡率升高(P<0.05),细胞SEPT2 mRNA与蛋白表达、细胞活力、集落形成率、迁移率、侵袭数、裸鼠肿瘤体积与质量降低(P<0.05);与miR-545-3p微泡组相比,miR-545-3p+SEPT2过表达微泡组细胞凋亡率降低(P<0.05),细胞SEPT2 mRNA与蛋白表达、细胞活力、集落形成率、迁移率、侵袭数、裸鼠肿瘤体积与质量升高(P<0.05)。结论:超声微泡介导miR-545-3p可通过降低SEPT2表达而减弱乳腺癌细胞增殖、迁移与侵袭,促使其凋亡,并抑制裸鼠移植瘤生长,最终抑制乳腺癌细胞恶性进展。Objective:To study the mechanism of miR-545-3p mediated by ultrasound microbubbles in inhibiting the malignant progression of breast cancer by regulating septin 2(SEPT2).Methods:The expression of miR-545-3p and SEPT2 in human normal breast epithelial cells MCF-10A,human breast cancer cells MCF-7,HS-578T and HCC-1937 cells were detected by real-time fluorescent quantitative PCR and Western blot.MCF-7 cells were cultured in vitro and randomly grouped into control group,empty microbubble group,miR-545-3p microbubble group,and miR-545-3p+SEPT2 overexpression microbubble group,after grouping and processing,real-time fluorescence quantitative PCR and immunoblotting experiments were applied to detect the expression of miR-545-3p and SEPT2 of cells in each group.CCK-8,colony formation assay,and flow cytometry were applied to detect cell proliferation and apoptosis in each group.Cell scratch and Transwell invasion tests were applied to detect cell migration and invasion in each group.Nude mice were subcutaneously inoculated with MCF-7 cells on the abdominal wall to construct their transplanted tumor model and grouped using the same method.After grouping,the tumor volume and weight of each group of nude mice were measured.Results:Compared with MCF-10A cells,the expression of miR-545-3p in MCF-7,HS-578T,and HCC-1937 cells decreased(P<0.05),the expression of SEPT2 mRNA and protein increased(P<0.05).Compared with the control group,the miR-545-3p expression and apoptosis rate in the miR-545-3p microbubble group increased(P<0.05),the cell SEPT2 mRNA and protein expression,cell viability,colony formation rate,migration rate,invasion number,and tumor volume and weight in nude mice reduced(P<0.05).Compared with the miR-545-3p microbubble group,the cell apoptosis rate in the miR-545-3p+SEPT2 overexpression microbubble group decreased(P<0.05),the cell SEPT2 mRNA and protein expression,cell viability,colony formation rate,migration rate,invasion number,and tumor volume and weight in nude mice increased(P<0.05).Conclusion:Ultrasound m
