摘 要:目的:基于膜相关酪氨酸/苏氨酸蛋白激酶1(protein kinase membrane associated tyrosine/threonine 1,PKMYT1)在乳腺癌中的差异表达探索PKMYT1对乳腺癌生物学行为的影响,以及经生存分析预测PKMYT1对乳腺癌预后的影响。方法:经生物信息学分析PKMYT1在泛癌及乳腺癌中的表达情况,预测PKMYT1对乳腺癌生物学行为的影响。并通过Kaplan-Meier Plotter数据库分析PKMYT1与乳腺癌预后的相关性。细胞实验验证PKMYT1对乳腺癌细胞系MCF-7、MDA-MB-231生物学行为的影响,通过细胞计数试剂盒(CCK-8)和Transwell分析,评估了PKMYT1对MCF-7和MDA-MB-231细胞增殖和侵袭能力的影响,且用流式细胞术检测PKMYT1对乳腺癌细胞周期及细胞凋亡的影响。结果:通过生物信息学分析PKMYT1在多种肿瘤样本中呈现高表达,其中包括乳腺癌,且具有统计学差异(P<0.05);经分析预测PKMYT1的表达与乳腺癌的细胞周期、DNA损伤及修复、侵袭、增殖及凋亡等生物学行为均呈正相关关系;细胞实验结果发现,敲低PKMYT1后可抑制MCF-7、MDA-MB-231细胞的增殖和侵袭能力,且在细胞周期的检测中G_(0)/G_(1)期增加、S+G_(2)/M期减少表明抑制了乳腺癌细胞的增殖,并且促进其发生凋亡。生存分析显示PKMYT1在乳腺癌中HR>1提示为高风险因素,且P<0.05表明PKMYT1在乳腺癌中与不良预后密切相关。结论:PKMYT1在乳腺癌样本中表达上调且与不良预后密切相关,PKMYT1或可通过调节细胞周期而加速乳腺癌的恶性进展,PKMYT1有望成为乳腺癌潜在的治疗靶点。Objective:To clarify the effect of protein kinase membrane associated tyrosine/threonine 1(PKMYT1)on the biological behavior of breast cancer based on the differential expression of PKMYT1 in breast cancer,and to predict the effect of PKMYT1 on the prognosis of breast cancer by survival analysis.Methods:The expression of PKMYT1 in pancarcinoma and breast cancer was analyzed by bioinformatics to predict the effect of PKMYT1 on biological behavior of breast cancer.The Kaplan-Meier Plotter database was used to analyze the correlation between PKMYT1 and the prognosis of breast cancer.The effect of PKMYT1 on the biological behavior of breast cancer cell lines MCF-7 and MDA-MB-231 was verified by cell assay,and analyzed by CCK-8 and Transwell.The effects of PKMYT1 on the proliferation and invasion of MCF-7 and MDA-MB-231 cells were evaluated,and the effects of PKMYT1 on the cell cycle and apoptosis of breast cancer cells were detected by flow cytometry.Results:By bioinformatics analysis,PKMYT1 was highly expressed in a variety of tumor samples,including breast cancer,and the difference was statistically significant(P<0.05).It was predicted that PKMYT1 expression was positively correlated with the cell cycle,DNA damage and repair,invasion,proliferation and apoptosis of breast cancer.Cell experiment results showed that PKMYT1 knockdown could inhibit the proliferation and invasion of MCF-7 and MDA-MB-231,and the increase of G_(0)/G_(1) phase and decrease of S+G_(2)/M phase in the detection of cell cycle indicated that the proliferation of breast cancer cells was inhibited and apoptosis was promoted.Survival analysis showed that PKMYT1's HR>1 in breast cancer was a high risk factor,and P<0.05 indicated that PKMYT1 was closely associated with poor prognosis in breast cancer.Conclusion:PKMYT1 expression is up-regulated in breast cancer samples and is closely associated with poor prognosis.PKMYT1 may accelerate the malignant progression of breast cancer by regulating cell cycle.PKMYT1 is expected to become a potential therape
