摘 要:目的:优化广西钩藤总黄酮的超声波辅助提取工艺,并检测其体外抗氧化活性。方法:采用UV法检测钩藤总黄酮含量;以总黄酮提取率为考察指标,采用正交设计法优化钩藤总黄酮的超声波辅助提取工艺条件;利用DPPH法检测乙醇提取物清除DPPH的活性。结果:采用超声波辅助提取法提取两次,超声功率360 W,最佳提取工艺为60倍量60%乙醇、超声提取30 min、提取温度70℃;在0.05~0.25 mg/mL范围内,随总黄酮提取物浓度的增加,提取物对DPPH的清除率增大;当提取物浓度为0.25 mg/mL时,柳州、梧州、钦州、玉林和桂林等5个产地的钩藤总黄酮对DPPH的清除率均在85%以上,分别为92.0%、91.6%、86.9%、90.8%、85.7%和97.3%。结论:该提取工艺稳定可行,可作为钩藤总黄酮的提取工艺;钩藤黄酮醇提取物对DPPH具有清除作用,可为钩藤黄酮的深入开发提供参考。Objective:To optimize the ultrasonic assisted extraction process of total flavonoids from Guangxi gambir plant and detect their in vitro antioxidant activity.Method:UV method was used to detect the total flavonoid content in gambir plant;Using the extraction rate of total flavonoids as the evaluation index,the orthogonal design method was used to optimize the ultrasonic assisted extraction process conditions of total flavonoids in Hook Vine;Use DPPH method to detect the activity of ethanol extract in clearing DPPH.Result:The ultrasound assisted extraction method was used twice,with an ultrasound power of 360W.The optimal extraction process was 60 times the amount of 60%ethanol,ultrasound extraction for 30 minutes,and extraction temperature of 70℃;Within the range of 0.05~0.25 mg/mL,as the concentration of total flavonoid extract increases,the clearance rate of the extract towards DPPH increases;When the concentration of the extract was 0.25 mg/mL,the total flavonoids of gambir plant from five production areas including Liuzhou,Wuzhou,Qinzhou,Yulin,and Guilin all had a clearance rate of over 85%for DPPH,which were 92.0%,91.6%,86.9%,90.8%,85.7%,and 97.3%,respectively.Conclusion:The extraction process is stable and feasible,and can be used as the extraction process for the total flavonoids of gambir plant;The alcohol extract of Houttuynia cordata has a clearing effect on DPPH,which can provide reference for the in-depth development of Houttuynia cordata flavonoids.
