摘 要:以经过康宁木霉菌LZ51(Trichoderma koninggii)发酵处理得到的发酵桑黄为原料,总多糖提取率为指标,采用正交试验与单因素实验相结合的方法,考察浸提过程中提取剂用量、提取时间及提取温度对提取率的影响,确定最佳提取工艺。结果表明:以水为提取剂、用量为45 mL/g, 100℃下浸提2 h,总多糖提取率最高,为48.44 mg/g。而同批次未经发酵前处理的桑黄,在相同条件下浸提,总多糖提取率仅为29.36 mg/g,远低于发酵桑黄总多糖提取率。由此可见,微生物发酵前处理可显著提高桑黄总多糖提取率。With the Phellinus igniarius obtained by LZ51(Trichoderma koninggii)as the raw material,the total polysaccharide extraction rate as the inspection index,the orthogonal experiment and single factor experiment were used to investigate the effects of the amount of extracting agent,the extraction time and the extraction temperature on the extraction rate during the reflux extraction process.The results showed that the highest extraction rate of total polysaccharide was 48.44 mg/g when water was used as extraction agent,the dosage was 45 mL/g,and the extraction time was 2 h at 100℃.The total polysaccharide extraction rate of the same batch of Phellinus igniarius without fermentation treatment was only 29.36 mg/g.It could be seen that the pretreatment of microbial fermentation can significantly improve the extraction rate of total polysaccharide from Phellinus igniarius.
