摘  要:代谢型谷氨酸受体(mGluRs)是中枢谷氨酸能系统中重要的受体之一,其广泛参与调控突触传递、突触可塑性、神经兴奋性和抑制性平衡等生理过程。mGluRs在介导胶质瘤的发生和发展中起重要作用,其介导的细胞内信号通路参与胶质瘤的进展、侵袭和复发过程,因此mGluRs作为胶质瘤的潜在药物靶点越来越受到关注。其中,mGluR1和mGluR3的体外药物阻断已被证明对于胶质瘤细胞具有抗细胞增殖和迁移的作用,mGluR3拮抗剂对于替莫唑胺有增加敏感性的作用,mGluR4和mGluR8可能成为治疗恶性胶质瘤的潜在靶点,但需要广泛的研究验证。未来应对不同类型的mGluRs在胶质瘤细胞中的作用进行深入探索,以为临床药物的研发提供理论依据。Metabotropic glutamate receptors(mGluRs)are one of the important receptors in the central glutamatergic system,widely involved in regulating physiological processes such as synaptic transmission,synaptic plasticity,neural excitability and inhibitory balance.mGluRs play an important role in mediating the occurrence and development of glioma.The intracellular signaling pathways mediated by mGluRs are involved in the progression,invasiveness,and recurrence of glioma,so they are increasingly recognized as potential drug targets for glioma.Among them,the in vitro pharmacological blockade of mGluR1 and mGluR3 has been shown to have anti-cell proliferation and migration effects on glioma cells,while mGluR3 antagonists have been shown to increase the sensitivity of temozolomide,mGluR4 and mGluR8 receptors could also be potential therapeutic targets for aggressive glioblastoma,but extensive studies and validation are needed.In the future,deep exploration should be conducted on the roles of different types of mGluRs in glioma cells in order to provide a theoretical basis for the development of clinical drugs.
