摘 要:目的 探讨具核梭杆菌(F.nucleatum)在口腔常用材料(钛、树脂义齿、聚苯乙烯)表面的聚集、黏附和生物膜形成特性,以及糖(蔗糖、乳糖、木糖醇)暴露对其的影响,旨在揭示F.nucleatum在不同口腔条件下的致病力。方法 将F.nucleatum在不同载体表面、不同糖暴露下厌氧培养2 h、4 h、6 h、8 h(黏附时期)、12 h、24 h、48 h、72 h、96 h(生物膜时期),采用结晶紫染色法检测F.nucleatum在不同条件下各时间点生物量,并计算黏附率;采用场发射扫描电子显微镜表征黏附时期的F.nucleatum形态。将F.nucleatum于不同材料(钛、树脂义齿、聚苯乙烯)表面、含有不同糖(蔗糖、乳糖、木糖醇)的培养基中厌氧培养48 h形成生物膜,将生物膜中细菌重悬后静置检测上清液吸光度值计算得到自聚集率。结果 0.1 m、0.5 m、1.5 m、3 m、4.0 m钛片、聚苯乙烯、树脂义齿表面F.nucleatum自聚集率分别为36.14%、53.76%、46.18%、45.08%、44.83%、50.22%;蔗糖、乳糖、木糖醇暴露后自聚集率分别为60.58%、43.36%、36.50%。F.nucleatum在0.1 m、0.5m、1.5 m、3 m钛片、树脂义齿表面于6 h达到最大黏附率,黏附率分别为98.00%、180.00%、177.00%、40.00%;4 m钛片、聚苯乙烯8 h黏附率分别为108.00%、548.00%。蔗糖、乳糖、木糖醇最大黏附率分别为370.00%、196.00%、144.00%、97.00%。所有条件下,F.nucleatum均在48 h达到最大生物膜生物量,生物膜成熟,72 h、96 h生物膜生物量减少,生物膜走向衰亡。结论 F.nucleatum在不同载体上、不同糖暴露下具有较好的聚集、黏附、形成生物膜能力。Objective To study the accumulation,adhesion and biofilm formation characteristics of Fusobacterium nucleatum(F.nucleatum)on the surface of commonly used oral materials(titanium,resin denture,polystyrene),and the effect of sugar(sucrose,lactose,xylitol)exposure on it.To reveal the pathogenicity of F.nucleatum in different oral conditions.Methods F.nucleatum was incubated anaerobically on different carrier surfaces and exposed to different sugars for 2 h,4 h,6 h,8 h(adhesion stage),12 h,24 h,48 h,72 h,96 h(biofilm stage).The biomass of F.nucleatum at different time points was detected by crystal violet staining method,and the adhesion rate was calculated.The morphology of F.nucleatum was characterized by field emission scanning electron microscopy.F.nucleatum was incubated anaerobically on the surface of different materials(titanium,resin denture,polystyrene)and in the medium containing different sugars(sucrose,lactose,xylitol)for 48h to form a biofilm.The absorbance value of the supernatant of the biofilm was measured by standing after the bacteria were suspended.The self-aggregation rate was calculated.Results The autoaggregation rates of F.nucleatum on 0.1 m,0.5 m,1.5 m,3 m,4.0 m titanium,polystyrene and resin denture surfaces were 36.14%,53.76%,46.18%,45.08%,44.83%,50.22%,respectively.The autoaggregation rates of F.nucleatum in sucrose,lactose and xylitol were 60.58%,43.36%and 36.50%,respectively.F.nucleatum reached the maximum adhesion rate of 98%,180%,177%,40%at 6 hours on 0.1 m,0.5 m,1.5 m,3 m,titanium disks and resin denture surfaces,respectively.The adhesion rates on 4m titanium sheet and polystyrene surface for 8h were 108%and 548%,respectively.The maximum adhesion rates of sucrose,lactose and xylitol were 370%,196%,144%and 97%,respectively.F.nucleatum reached the maximum biomass of biofilm at 48h and biofilm matured,but biofilm biomass decreased at 72 h and 96 h and biofilm declined.Conclusion F.nucleatum has good ability to aggregate,adhere and form biofilms on different vectors and at different sugar
