甘蓝型油菜自交不亲和性基因遗传分析与初定位

摘  要:自交不亲和性是利用油菜(Brassica sp.)杂种优势的途径之一。自交不亲和系杂种有诸如没有不育胞质存在的负效应、恢复源广泛、可获得高质量F1代种子等优点,因此研究调控自交不亲和性的基因对利用油菜杂种优势具有重要意义。以甘白(甘蓝型油菜×白菜型油菜,B.napus×B.rapa)杂交选育的甘蓝型油菜自交不亲和系No.234为研究对象,通过遗传规律分析发现No.234不同遗传背景的恢复系的自交不亲和性分别受1、2对隐性基因(BnSI1、BnSI2)控制;利用AFLP(扩增片段长度多态性)标记和BSA-seq(极端性状混池重测序)将自交不亲和基因BnSI1初步定位在A07染色体2 Mb区间内。该研究为进一步定位、克隆BnSI1基因及后续的功能研究提供研究基础。As one of the ways to exploit the heterosis in rape(Brassica sp.),the self-incompatibility line has no negative effect on the existence of sterile cytoplasm,has a wide range of recovery sources,can obtain high-quality F1 generation seeds,etc.Therefore,it is important to study relative genes regulating self-incompatibility to exploit the heterosis in rape.In this study,the self-incompatibility line No.234,a B.napus selected from a cross between B.napus and B.rapa,was used to study the genetic pattern.It shows that the self-incompatibility of No.234 was controlled by one and two pairs of recessive genes(BnSI1 and BnSI2),respectively,for recovery lines of different genetic backgrounds.AFLP(amplified fragment length polymorphism) markers and BSA-seq(bulked-segregant analysis sequencing) were used to initially locate the self-incompatibility gene BnSI1 in the 2 Mb interval of chromosome A07.This study may lay the foundation for further localization and cloning of BnSI1 gene,and subsequent functional studies.
