基于生物信息学和实验探讨PDXK在肝脏肝细胞癌的表达及临床价值

摘  要:目的:基于生物信息学分析和实验验证探讨吡哆醛激酶(PDXK)在肝脏肝细胞癌(LIHC)中的表达及其临床价值。方法:利用TIMER、UALCA、HPA、STRING、DAVID数据库分析PDXK在LIHC和正常组织中的mRNA和蛋白表达、PDXK与免疫细胞浸润、预后、不同临床特征、癌症分期分级及甲基化的相关性;并构建PDXK相关蛋白网络和进行功能富集分析;利用倒置显微镜、CCK8法分别检测肝癌细胞Bel7404敲低PDXK后其细胞生长形态和细胞存活率的变化。结果:PDXK在泛癌中高表达,在LIHC中PDXK的mRNA和蛋白表达显著高于正常组织,PDXK与LIHC患者的不同临床病理特征、癌症分期分级、预后生存及免疫浸润密切相关,PDXK及其相关基因的功能主要富集参与3个信号通路、4个分子功能、4个细胞组分和1个生物学过程,敲低PDXK可抑制Bel7404细胞增殖、降低细胞存活率。结论:PDXK是促进肝癌发生发展的关键风险因子,PDXK在肝癌中高表达且与临床预后、肝癌细胞存活呈负相关。Objective:To investigate the expression and clinical value of pyridoxal kinase(PDXK)in liver hepatocellular carcinoma(LIHC)based on bioinformatics analysis and experimental validation.Methods:The TIMER,UALCA,HPA,STRING,and DAVID databases were used to analyze the mRNA and protein expression of PDXK in LIHC and normal tissues,as well as the correlation between PDXK and immune cell infiltration,prognosis,different clinical features,cancer staging and grading,and methylation.And construct a PDXK related protein network and conduct functional enrichment analysis.Inverted microscope and CCK8 method were used to detect cell growth morphology and cell survival rate of Bel7404 liver cancer cell after knocking down PDXK.Results:PDXK is highly expressed in pan cancer,and the mRNA and protein expression of PDXK is significantly higher in LIHC than in normal tissues.PDXK is closely related to different clinical and pathological characteristics,cancer staging and grading,prognosis,and immune infiltration of LIHC patients.The function of PDXK and its related genes is mainly enriched and involved in three signaling pathways,four molecular functions,four cell components,and one biological process.Knocking down PDXK can inhibit the proliferation of Bel7404 cells and reduce cell survival rate.Conclusion:PDXK is a key risk factor promoting the occurrence and development of liver cancer.PDXK is highly expressed in liver cancer and negatively correlated with clinical prognosis and liver cancer cell survival.
