摘 要:目的优化铜藻岩藻多糖的匀浆提取工艺,并测定其抗氧化活性。方法以岩藻多糖的提取率为指标,基于单因素试验的基础上,采用响应面法对铜藻中岩藻多糖的提取工艺进行优化,并对其抗氧化活性进行验证。结果最佳提取工艺条件为提取时间6 min、提取温度94℃、液料比34 mL·g^(-1)和提取次数2次。在此条件下,多糖的提取率为14.76%。抗氧化活性研究结果表明,铜藻岩藻多糖在质量浓度为0.2~10.0 mg·mL^(-1)范围内有一定的抗氧化活性,对DPPH自由基、ABTS+自由基清除率的IC_(50)分别为0.22、0.41 mg·mL^(-1)。结论该提取工艺高效,可为铜藻岩藻多糖的制备及应用提供理论依据。Objective To optimize the homogenization extraction process of Sargassum horneri fucoidan and determine its antioxidant activity.Methods Based on the extraction yield of fucoidan and single factor experiments,response surface methodology was used to optimize the extraction process of fucoidan from Sargassum horneri,and its antioxidant activity was verified.Results The optimal extraction process conditions were extraction time of 6 min,extraction temperature of 94℃,liquid-solid ratio of 34 mg·mL^(-1),and extraction times of 2 times.Under these conditions,the yield of fucoidan was 14.76%.The results of antioxidant activity research indicate that Sargassum horneri fucoidan has certain antioxidant activity in the mass concentration range of 0.2~10.0 mg·mL^(-1),with IC 50 of 0.22 mg·mL^(-1)and 0.41 mg·mL^(-1)for DPPH and ABTS free radical scavenging rates,respectively.Conclusion The extraction process was efficient and can provide a theoretical basis for the preparation and application value of Sargassum horneri fucoidan.
