高原缺氧环境对雌性大鼠生殖系统的影响

摘　　要：探讨高原缺氧环境暴露对雌性大鼠生殖系统的影响。将20只雌性Wistar大鼠进行生理同步化后随机分为平原组与高原组,平原组置于兰州(海拔1500 m),高原组运输至青海玉树巴塘(海拔4010 m)后高原缺氧环境暴露14 d。实验过程中记录大鼠体质量和发情周期变化,取大鼠各个组织器官计算器官指数,检测血清、卵巢和子宫组织生殖激素水平和氧化应激水平的变化,HE染色观察卵巢和子宫组织形态学的变化,蛋白质印迹法检测卵巢组织中激素受体蛋白的表达量。与平原组相比,高原组缺氧暴露后大鼠体质量明显下降,发情周期紊乱,在血清、卵巢和子宫组织中氧化应激水平显著上调且生殖激素水平紊乱。HE染色结果显示,平原组大鼠卵巢和子宫的形态结构完整,高原组卵巢和子宫组织结构遭到破坏。卵巢组织中激素受体蛋白含量较平原组减少。高原缺氧会导致雌性大鼠生殖损伤。To investigate the effects of exposure to plateau hypoxia on the reproductive system of female rats.Twenty female Wistar rats were physiologically synchronized and randomly divided into Plain group and Plateau group.The Plain group was placed in Lanzhou(1500 m above sea level),while the Plateau group was transported to Batang,Yushu,Qinghai Province(4010 m above sea level)and then exposed to plateau hypoxia for 14 days.During the experiment,the body weight and estrous cycle changes were recorded,the organ index was calculated from each tissue and organ.The changes in reproductive hormone levels and oxidative stress levels in serum,ovary and uterus tissues were detected,HE staining was used to observe the morphology of ovary and uterus tissues,and the expression of hormone receptor proteins in ovary tissues was detected by protein blotting method.Compared with the Plain group,rats in the Plateau group showed significant weight loss,estrous cycle disruption,up-regulation of oxidative stress and reproductive hormone levels in serum,ovary and uterus tissues,and HE staining showed that the morphology and structure of ovary and uterus in the Plateau group were intact,whereas the struc ture of ovary and uterus in the Plateau group was damaged.The amount of hormone receptor proteins in the o varian tissue was reduced compared with that in the Plateau group.Hypoxia in the Plateau group caused reproductive damage in female rats.
