摘 要:空气污染是肝细胞癌(HCC)发病的重要原因。本文从癌症基因组图谱(TCGA)数据库、国际肿瘤基因组协作组(ICGC)数据库和CTD数据库下载的三个基因集,得到HCC在发生发展过程中受空气污染影响的共同关联基因。利用DAVID数据库、Metascape在线平台进行GO和KEGG富集分析,STRING数据库分析并绘制PPI蛋白互作网络图,采用单因素Cox分析筛选出对肝细胞癌预后有预测功能的关键基因。研究结果表明:在119个共同关联基因中筛选出8个预后关键基因,相对于肝正常组织,关键基因在肝癌肿瘤组织中mRNA及蛋白表达水平呈现高表达,在生存分析中显示预后不良,对不同临床特征和预后生存具有重要指导作用。8个预后关键基因可用作受空气污染影响的HCC潜在标志物,为肝癌潜在诊断和预后生物标志物的识别提供理论依据。Air pollution is an important cause of hepatocellular carcinoma(HCC).In this study,three gene sets were downloaded from the Cancer Genome Atlas(TCGA)database.the International T umor Genome Collaboration(ICGC)database and the CTD database,and the common associated genes afected by air pollution in the development and development of HCC were obtained.DAVID database and Metascape online platform were used for GO and KEGG enrichment analysis,String database was analyzed and PPI protein interaction network map was drawn,and unifactor Cox analysis was used to screen out key genes that can predict the prognosis of hepatocellular carcinoma.The study showed that among 119 corassociated genes,8 key prognostic genes were screened out.Compared with normal liver tissues.mRNA and protein expression levels of key genes were highly expressed in liver cancer tumor tissues,indicating poor prognosis in survival analysis,and playing an important role in different clinical characteristics and prognostic survival.Eight key genes can be used as potential markers of air pollution in HCC,providing a new theoretical basis for the identification of potential diagnostic and prognostic biomarkers of hepatocellular carcinoma.
