摘 要:目的 利用UPLC-Q-Exactive Orbitrap-MS法对三化汤化学成分进行鉴定。方法 分析采用Waters CORTECS T3色谱柱(2.1 mm×150 mm, 1.6μm);流动相乙腈-0.2%甲酸,梯度洗脱;体积流量0.2 mL/min;柱温30℃;检测波长260 nm;加热电喷雾离子源(HESI);正负离子监测模式;质量扫描范围m/z 100~1 200。采用Xcalibur 2.1软件并结合文献报道、质谱数据对化学成分进行定性鉴别。结果 从三化汤中共鉴定出166个化合物,包括黄酮类、蒽醌及蒽酮类、香豆素类、木脂素类、生物碱类等,其中对照品指认的成分有30种。结论 该方法可实现对三化汤化学成分的快速鉴别,为后续探究该经典名方药效物质基础、建立其质量标准奠定基础。AIM To identify the chemical constituents in Sanhua Decoction by UPLC-Q-Exactive Orbitrap-MS.METHODS The analysis was performed on a 30℃thermostatic Waters CORTECS T3 column(2.1 mm×150 mm,1.6μm),with the mobile phase of acetonitrile-0.2%formic acid flowing at 0.2 mL/min in a gradient elution manner,and the detection wavelength was set at 260 nm.The heated electrospray ionization source(HESI)was adopted in positive and negative ion monitoring mode,and the mass scan range was m/z 100-1200.The chemical constituents were qualitatively identified by Xcalibur2.1 software combined with literature reports and mass spectrometry data.RESULTS A total of 166 compounds were identified in Sanhua Decoction,including flavonoids,anthraquinones and anthrones,coumarins,lignans and alkaloids,of which 30 were identified by reference substances.CONCLUSION The method established in this paper can realize the rapid identification of the chemical components of Sanhua Decoction,and lay a foundation for the follow-up study of pharmacodynamic material basis and establishment of quality standard for this famous classical formula.
