摘  要:采用RP-HPLC法,以乙腈(A)-水(B)为流动相梯度洗脱,检测波长210 nm,柱温25℃,流速1 mL·min^(-1);在单因素实验基础之上,采用Box-Behnken响应面分析法优化杜鹃醇的最优工艺条件。杜鹃醇在0.1~2.5μg(r=0.9998)内具有良好的线性关系,平均加样回收率为97.90%,RSD=1.38%,榜留丽叶中杜鹃醇含量在7.02~23.23 mg/g间。在最佳工艺条件下:乙醇浓度为90%、料液为1∶50、温度为90℃、提取3次、每次2 h,杜鹃醇含量达8.17 mg/g。本文所筛选的RP-HPLC检测方法可作为苗药榜留丽的质量控制方法。Using RP-HPLC method,using acetonitrile(A)-water(B)as mobile phase gradient elution,detection wavelength of 210 nm,column temperature of 25℃,flow rate of 1 mL·min^(-1),based on single factor experiment,using Box-Behnken response surface analysis method,the optimal process conditions of Rhododendol were optimized.Box-Behnken response surface analysis method was used to optimize the optimal process conditions of Rhododendron on the basis of single factor experiment.The linear range of Rhododendrol was 0.1~2.5μg,the sample recovery rate was 97.90%,and RSD was 1.38%.The best extraction process for Rhododendol was as follows:ethanol concentration was 90%,solid-liquid ratio was 1∶50,extraction temperature was 90℃,extraction times were three times each time was 2 h,under this condition,the rhododendron content was 8.17 mg/g.This RP-HPLC detection method and extraction process provided a reference for the rational use of the folk medicine Rhododendron delavayi Franch.
