摘 要:目的制备蝮蛇特异性抗体,为进一步研发抗蛇毒血清提供理论依据。方法以蝮蛇蛇毒蛋白作为抗原,通过免疫新西兰兔制备蝮蛇特异性抗体,应用间接ELISA法检测其效价,Western blot检测其特异性。结果成功制备了蝮蛇特异性抗体血清,其效价可达1∶100000,Western blot检测抗体对蝮蛇蛇毒具有专一性。结论本研究成功制备了蝮蛇特异性抗体,推进了抗蝮蛇毒血清的研发。Objective The preparation of specific antibodies to Agkistrodon halys,provided theoretical basis for further research and development of antivenoms.Methods Using the venom protein of Agkistrodon halys as antigen,the specific antibody of Agkistrodon halys was prepared by immunizing New Zealand rabbits.Its titer was detected by indirect ELISA and its specificity was detected by Western blot.Results The specific antibody serum of Agkistrodon halys was successfully prepared,and its titer was 1∶100000.Western blot analysis showed that the antibody was specific to Agkistrodon halys venom.Conclusion This study successfully prepared the specific antibody of Agkistrodon halys and promoted the research and development of anti-Agkistrodon halys venom serum.
