摘 要:目的研究一种运用改良真空冷冻干燥技术改进消毒剂杀菌试验中菌片制备的方法。方法使用改良真空冷冻干燥技术将大肠杆菌和白念珠菌固定在载体上制备真空冻干菌片,采用保存有效期监测试验、稳定性模拟试验及载体定量杀菌试验,对该真空冻干菌片保存有效期、稳定性以及是否影响消毒效果进行评价。结果-20℃保存条件下,载体类型为布片、钢片时,大肠杆菌和白念珠菌菌片保存有效期均为3个月;-80℃保存条件下,大肠杆菌和白念珠菌布片保存有效期为4个月,钢片稳定性最好,保存有效期为6个月。运输温度不高于25℃、运输时间不超过4h以及中和剂浸泡温度不高于25℃、浸泡时间不超过4h,真空冻干菌片的回收菌量仍能满足试验要求。分别使用常规干燥法和真空冻干法制备菌片,使用有效氯含量为150g/L的84消毒液作用3min,两种菌片制备方法中大肠杆菌的杀灭对数值均>3.00;使用有效氯含量为150g/L的84消毒液作用5min,两种菌片制备方法中白念珠菌的杀灭对数值均>3.00;使用过氧乙酸含量为100g/L的消毒液作用1min,两种菌片制备方法中大肠杆菌的杀灭对数值均>3.00;使用过氧乙酸含量为100g/L的消毒液作用3min,两种菌片制备方法中白念珠菌的杀灭对数值均>3.00。结论改良后的真空冻干技术可明显延长真空冻干菌片的保存时间,且能保持良好的稳定性。Objective To study a method of improving the preparation of bacteria tablet in disinfectant sterilization test by vacuum freeze-drying technology.Methods The improved vacuum freeze-drying technology was used to prepare the vacuum freeze-dried bacteria tablets by fixation of Escherichia coli and Candida albicans on the carrier.The storage period monitoring test,stability simulation test and carrier quantitative sterilization test were used to evaluate the storage period and stability of the vacuum freeze-dried bacteria tablets and whether they affected the disinfection effect.Results Under the storage condition of-20℃,the validity period of Escherichia coli and Candida albicans bacterial tablets was 3 months when the carrier type was cloth and steel tablets.Cloth tablets of Escherichia coli and Candida albicans were stored for 4 months at-80℃,while steel tablets had the best stability and were stored for 6 months.With the transportation temperature not higher than 25℃and transportation time not more than 4 h as well as the neutralizer immersion temperature not higher than 25℃and immersion time not more than 4 h,the recovered bacterial amount of vacuum lyophilized bacterial tablets could still meet the test requirements.The bacteria tablets were prepared by conventional drying method and vacuum freeze-drying method,respectively.When 84 disinfectant with an effective chlorine content of 150 g/L was used for 3 min,the logarithm of killing of Escherichia coli in the 2 methods was more than 3.00.When 84 disinfectant with an effective chlorine content of 150 g/L was used for 5 min,the logarithm of killing value of Candida albicans in the 2 preparation methods was more than 3.00.When 100 g/L peracetic acid disinfectant was used for 1min,the logarithm of Escherichia coli killing was more than 3.00 in the two preparation methods.Using 100 g/L peracetic acid disinfectant for 3 minutes,the logarithm of killing Candida albicans in the 2 preparation methods was more than 3.00.Conclusion The improved vacuum freeze-dryin
