摘  要:以富硒小麦全粉为原料,利用超声辅助纤维素酶法提取小麦硒多糖。以小麦硒多糖提取率为衡量指标,通过单因素实验和响应面法对小麦硒多糖提取工艺进行优化,并研究其对ABTS+的抗氧化活性。结果显示,小麦硒多糖的最佳提取工艺条件为料液比1:17(g:mL)、纤维素酶浓度0.75%、超声温度60℃、超声功率497 W、超声时间50 min。在此条件下小麦硒多糖提取率为38.24%,硒含量为0.093 mg/kg。体外抗氧化活性研究表明,其对ABTS^(+)自由基清除率IC_(50)为4.56 mg/mL。Selenium polysaccharides were extracted from selenium-rich whole wheat flour using ultrasound-assisted cellulase method.The extraction process of wheat selenium polysaccharides was optimized by single-factor experiment and response surface methodology,with the extraction rate of selenium polysaccharides as the evaluation index.The antioxidant activity of wheat selenium polysaccharides against ABTS+was also studied.The results showed that the optimal extraction conditions for wheat selenium polysaccharides were a solid-liquid ratio of 1:17(g:mL),cellulase concentration of 0.75%,ultrasound temperature of 60℃,ultrasound power of 497 W,and ultrasound time of 50 minutes.Under these conditions,the extraction rate of wheat selenium polysaccharides was 38.24%,and the selenium content was 0.093 mg/kg.The vitro antioxidant activity study showed that the IC_(50) value of wheat selenium polysaccharides for ABTS^(+) radical scavenging was 4.56 mg/mL.
