双水相萃取茯苓多糖的工艺优化及其抗氧化活性评价

摘  要:采用聚乙二醇(PEG)-硫酸铵双水相体系萃取茯苓多糖。通过单因素试验考察和Box-Behnken响应面试验优化萃取工艺条件,得到优化工艺参数为聚乙二醇相对分子质量4000,聚乙二醇质量分数28.46%,硫酸铵质量分数20.60%,此时多糖的萃取率为97.57%。在此条件下,测定茯苓多糖对DPPH·和·OH的清除能力。结果表明茯苓多糖具有体外抗氧化活性,随着茯苓多糖质量浓度的提高,其抗氧化活性增强。A Polyethylene glycol(PEG)-ammonium sulfate aqueous two-phase system was employed for the extraction of Poria polysaccharide.The extraction process conditions were optimized through single-factor experiments and Box-Behnken response surface methodology,and the optimized parameters inclueded a polyethylene glycol molecular weight of 4000,a PEG mass fraction of 28.46%,and an ammonium sulfate mass fraction of 20.60%.Under these conditions,the extraction yield of Poria polysaccharide was 97.57%.Additionally,the determination of the scavenging ability of Poria polysaccharides on DPPH·and·OH.The results showed that Poria polysaccharide had antioxidant activity in vitro,which increased with increasing concentration of Poria polysaccharide.
