摘　　要：目的探讨基底前脑Nestin阳性神经元是否投射到内侧缰核、基底外侧杏仁核、腹侧被盖区和中脑脚间核及其规律。方法将36只SD大鼠编号后使用随机数字表分为4组,每组9只,分别在内侧缰核(MHb)、基底外侧杏仁核(BLA)、腹侧被盖区(VTA)和中脑脚间核(IP)注射逆行示踪剂快蓝(FB),5 d后取材,用免疫荧光染色观察基底前脑处Nestin、ChAT阳性神经元的投射情况。结果基底前脑可投射至内侧缰核、基底外侧杏仁核,但与腹侧被盖区、中脑脚间核并无投射环路,其中基底前脑-内侧缰核投射环路中约有14.5%由Nestin阳性神经元发出,这两个脑区之间的胆碱能环路中约有26.9%为Nestin亚型;基底前脑-基底外侧杏仁核投射环路中约有5.19%为Nestin阳性神经元,两脑区之间的胆碱能环路中有16.7%为Nestin亚型。结论除已有报道的脑区外,Nestin阳性神经元主要与内侧缰核形成神经环路,也少量投射至基底外侧杏仁核,可能与恐惧记忆、情绪等的调控有关。Objective To investigate whether Nestin-positive neurons in the basal forebrain project to the medial habenular nucleus,basolateral amygdala,ventral tegmental area,and interpeduncular nucleus,as well as the regularity of such projections.Methods A total of 36 SD rats were numbered and randomly divided into four groups using a random number table,with 9 rats in each group.Retrograde tracer fast blue(FB)was injected into the medial habenular nucleus(MHb),basolateral amygdala(BLA),ventral tegmental area(VTA),and interpeduncular nucleus(IP)respectively.After 5 days,the brains were collected,and the projection of nestin and ChAT-positive neurons in the basal forebrain was observed using immunofluorescence staining.Results The basal forebrain projects to the medial habenular nucleus and basolateral amygdala,but there was no projection circuit to the ventral tegmental area or interpeduncular nucleus.About 14.5%of the projection pathway from the basal forebrain to the medial habenular nucleus originates from Nestin-positive neurons,and about 26.9%of the cholinergic circuits between these two brain regions was of the nestin subtype.Additionally,about 5.19%of the projection pathway from the basal forebrain to the basolateral amygdala was composed of Nestin-positive neurons,and 16.7%of the cholinergic circuits between these two brain regions was of the nestin subtype.Conclusion Apart from the previously reported brain regions,Nestin-positive neurons primarily form neural circuits with the medial habenular nucleus and also project to the basolateral amygdala to a lesser extent.These neural circuits may be involved in the regulation of fear memory,emotions,and other related functions.
